The non-Hodgkin’s lymphomas(NHLs) are groups of lymph proliferative malignant diseases, and represent in the top ten of common malignant cancers. Three types of NHLs were divided: the indolent lymphoma, the aggressive lymphoma and the highly aggressive lymphoma. The indolent lymphoma accounts for about 25%-40% of all NHL diseases.Because the indolent lymphomas have a very secretive beginning and a very slow pathological process, the patients with the indolent lymphoma cannot discover their illness until the terminally stage. The conventional treatments rely on the CVP and CHOP chemotherapy combined with radiotherapy mainly, but with poor treatment effect. Patients with the indolent lymphomas usually get eased but not curable. Besides, the indolent lymphoma has highly recurrence rate. The treatment effects will be shorten and weaken after the recurrence. Only the patients with early-stage indolent lymphoma can get a radical curation by radiotherapy clinically. Therefore, how to detect the early-stage indolent lymphoma and in the most important part in improving the cure rate and survival rate for patients.Objective: To investigate the methylation state of SHP-1 and DAPK gene promoter and to evaluate its significance in indolent B-cell lymphoma. Methods:Specimens of PBMC from indolent B-cell lymphoma and specimens of PBMC from health individuals were studied, unmethylation-specific PCR and methylation-specific PCR were designed and adopted to detect the methylation state of SHP-1 gene promoter. Results: The frequency of methylation in SHP-1gene promoter in indolent B-cell lymphoma were high, however, specimens of health individuals showed on mythelation in SHP-1 and DAPK gene promoter(p<0.01). Conclusion: The detection rate of SHP-1 and DAPK methylation in indolent B-cell lymphoma was high, hypermethylation of SHP-1and DAPK gene promoter in PBMC can be used as a good molecular marker for the diagnosis of indolent B-cell lymphoma.
