| Purpose:To explore the influence of yang-warming and fluid retention-resolving therapy on rat vascular endothelial growth factor to reveal some of its possible mechanism on treating water disease.Methods:Using the digitally random methods to divides the adult Wistar rats into four groups after adaptive feeding a week, blank group, model group, the yang-warming and fluid retention-resolving group and wiping off the cassia twig fluid retention-resolving group, each group with ten rats(five male and five female per group). Injecting 1% carrageenan into pleura to establish the model of pleural effusion. After the model was established for 24h, model rats were subjected to imaging recognization by radiography. At 2h after the model was established, the yang-warming and fluid retention-resolving group and wiping off the cassia twig fluid retention-resolving group were dosed with the respective treatments by gavage once daily for 3 d. The other groups give regular feeding. All rats were sacrificed on the third day after the molding was established, collecting the blood, pleural effusion, visceral pleura and parietal pleura. Using enzme-linked immune sorbent assay(ELISA) determination to measuring the content of VEGF in the blood serum and pleural effusion. Using the electron microscope to observe the organizational structure about pleural. The quantitative data was statistical analysis by SPSS17.0.Results:(1) The content of VEGF in blank group, the yang-warming and fluid retention-resolving group and wiping off the cassia twig fluid retention-resolving group are lower than model group. There are statistically significant in multiple comparisons by SNK, P<0.05.(2) There are significant differences about VEGF between blank group, yang-warming and fluid retention-resolving group and wiping off the cassia twig flui d retention-resolving group control with the model group in blood serum, P<0. 05.(3) In the same group, there is no significant difference in VEGF of blood serum and pleural effusion, P>0.05.(4) There are some correlation between blood serum and pleural effusion abo ut VEGF through Pearson correlation analysis. The correlation coefficient is 0.4 65, P<0.05.(5) There are some correlation between blood serum and VEGF level of ple ural effusion through Pearson correlation analysis. The correlation coefficient is 0.465, P<0.05.(6) There is no significant difference in VEGF between yang-warming and f luid retention-resolving group and wiping off the cassia twig fluid retention-res olving group in multiple comparisons by SNK, P>0.05.(7) Rich microvilli on the surface of mesothelial cells, collagenous fiber and closely packed in visceral pleura and parietal pleura control the yang-warming and fluid retention-resolving therapy group and wiping off the cassia twig fluid retention-resolving group with the model group, and the collagenous fiber are closely packed between the yang-warming and fluid retention-resolving therapy group and wiping off the cassia twig fluid retention-resolving group but there are no significant difference.(8) Compared with the blank group, the content of pleual effusion in other t hree groups is increased. Compared with the model group, the wiping off the cassia twig fluid retention-resolving group is decreased after given the medicine. There is no significance between the yang-warming and fluid retention-resolvi ng group and the wiping off the cassia twig fluid retention-resolving gro up.Conclusion:(1) The content of VEGF in blood serum and pleural effusion in rat about pleural effusion model is increased, this indicate VEGF has a intimate connection with the formation of pleural effusion.(2) The yang-warming and fluid retention-resolving therapy can reduce the expression of VEGF in blood serum and pleural effusion with rats in pleural effusion models, it may be one of the mechanism to treat the disease of retained fluid.(3) In the same group, there is some expression correlation between blood serum and pleural effusion on VEGF, and there is no significant difference on express, this indicate that when there is a small amount of pleural and it’s difficult to draw out, we can detect the VEGF in blood serum to help us diagnose.(4) There is no difference in contents of pleural effusion and the VEGF in blood serum and pleural effusion between the yang-warming and fluid retention-resolving group and wiping off the cassia twig fluid retention-resolving group, but compared with the wiping off the cassia twig fluid retention-resolving group its collagenous fiber is closely packed, it shows that the cassia twig may play some role in this formula. |
