Acer mono (Acer mono Maxim) is Aceraceae Maple tree the color leaves and an important ornamental plants,By space mutation may have taken place in morphology genetic variation,from the ornamental value for the cultivation of more varieties of Acer mono provide breeding material.In this experiment,the space mutation of Acer mono seedlings cultivated groups and individuals for the materials , Chlorophyll and morphological of the comprehensive clustering. Using RAMP( random amp lified microsatellite polymorphism) markers of their groups genomic DNA were detected, Analysis of space radiation and the control group of seedlings population genetic differences,found whether was occurred by space after Acer mono the genomic DNA genetic variation.On this basis,Individuals from different leaves and roots of extracted genomic DNA,using RAMP(random amp lified microsatellite polymorphism)markers and SSR(simple sequence repeats)markers prove genetic variation within individual,provide a theoretical basis for Acer mono for the space mutation breeding of new varieties of quality. The main results are as follows:Using RAMP(random amplified microsatellite polymorphism)markers 10 pairs of primer combinations on six groups(three treatment and three control)were amplified in 120 individuals with 360 bands,received a total of 256 polymorphic loci polymorphic loci and rate of 71.11%.RAMP marker found High polymorphism within populations,Radiation group more than the control groups .In six population,every population effective number of alleles ranged between 1.4086-1.6485, average 1.5331 ;every population Observed heterozygosity (Ho) values of change in the range of 0.2986-0.3765,averaging 0.3303;every population expected heterozygosity (He) value of the change in the range 0.3454-0.4362,an average of 0.3837,Illustrate the level of polymorphism among populations quite different.By the effective number of alleles detected(Ne)and expected heterozygosity(He)as a standard,the genetic diversity of radiation group sm3 the highest level of genetic diversity were the lowest ck3.Using RAMP( random amp lified microsatellite polymorphism)markers 10 pairs of primer amplified in 120 individuals with 360 bands,Calculated among individuals within a population genetic similarity GS and genetic distance D.In six populations ranged from 0.8724-0.9375,genetic distance between 0.0645-0.1365,Shows a higher degree of similarity and small genetic distance between groups.Space radiation between the three groups and the control of the three groups were relatively high similarity coefficients and genetic distance is relatively small,and space treatment groups and control treatment groups is relatively low similarity coefficients and genetic distance is relatively large.The six populations are clustered into two major clades by UPGMA method based on genetic distance.Space radiation group and the control of the genetic distance of six groups,between 0.0645-0.1365.Populations of three-time repetition of the radiation are clustered into one clade,three-time repetition of the comparison are clustered into the second clade.
