South of China is the native orgins of longan. The history of longan cultivating has been thousands of years. New cultivars have been increasing by selection, breeding and tissue culture. 50 longan cultivars from guangxi and other regions were studied by fluorescent AFLP.A fingerp-rinting has been constructed. Relationship and inheritance among parents have been analyzed and some new cultivars have been classified on molecular level. The main results were as follows.1. Different time of leaf picking for DNA extraction was studied.Young leaves have higher antioxidation, and contain less impurities and get more genetic DNA than mature leaves have.Using modified 3xCTAB can improve the quality of DNA remarkably.2.10 primer combinations with high polymorphism were selected from 64 primer combinations.569 bands were amplified totally among 50 cultivars, in which 475 bands were polymorphic and the polymorphic percentage is 83.5%.3. A fingerprinting containing 50 cultivars has been constructed by AFLP.4. UPGMA clustering method was carried out by soft wares-NTSYS package.50 accessions could be taxonomied into ten groups.
