OBJECTIVES: To study the relationship between the genetic mutations of MTHFR C677T, CBS T833C, and ischemic stroke in the people of Han nationality of Southern China. METhODS: Delennination of the MThFR C677T gene mutation by PCR-RFLP technique and the CBS T833C gene mutation by ARMS method in 74 patients with ischcnic stroke and 83 normal people for control. RESIJIFS: The frequency of MTHFR T homogenetic type and T allele (2.7% and 0.28) in ischemic group was higher than those in control group (1.2% and 0.21). With age boundary, there was significant difference (p<0.05) in the frequency of MTHFR genotypes and T allele between ischemic group and control group (age60), whereas there was no significant difference (p>O.O5) in these groups (age)60). The frequency of CBS C homogenetic type and C allele (13.5% and 0.44) in ischemic group was higicr than those in control group (6.0% and 0.33). The frequency distribution of MTHFR and CBS genotypes was corresponding with Hardy-Weinberg equilibrium. Logistic Regression analysis showed that together with the T allele in MTHFR and the C allele in CBS were related to ischcmic stroke (p<0.05). The odds ratios (OR) of the T allele in MTHFR C677T and the C allele in CBS T333C were 1.74 (95%C1 1.06-2.86) and l.73(95%CI 1.07-2.81) respectively. CON(I.IiSIONS: I. Genetic mutations of MTI-1FR C677T and CBS T833C are found in the people of Han nationality of Southern China. The frequency distribution of MTHFR C677T and CBS T833C genotypes is corresponding with Hardy-Weinberg equilibrium. 2. The age boundary exiting in genetic mutation of MTHFR C677T suggests that gene MTHFR may be one of the sensitive genes in ischcinic stroke under 60. 3. Wc conlirni that the Gcnctic mutations of MTHFR C677T, CBS T833C. arc related with the ischcmic stroke in the people of Han nationality of Southern China. The gcnetic mutations of MTHFR C677T and CBS T833C may be genetic factors for ischcmic stroke.
