| Premature delivery refers to the gestation within 28-37 completed weeks.As premature infant is lower in weight and the development of organs is immature comparatively and the viability of the newborns is low, so it is the leading cause of neonatal mortality. Even though they can survive, they are suffered by intelligence impairment or neural sequelae, so the diagnosis and treatment of preterm delivery is very important in improving the population quality. The study of human parturition mechanism has an important role in the diagnosis and treatment of premature delivery. CRH is synthesized and secreted mostly from placenta syncytiotrophoblast cells in gestation, and placental CRH is secreted into maternal circulation during second and third trimester of pregnancy. Some studies have showed, there are abnormal elevated maternal plasma CR11 levels in preterm delivery women,which indicates CR11 has a close correlation with human parturition. But the definite correlation of CR11 with preterm labor and its mechanism in human parturition is incompletely understood now.Our study will investigate what correlation plasma CR11 has with preterm labor and its probable mechanism.This experiment is composed of three parts. Firstly,we measure the level of plasma CR11 content in different pregnancy weeks,to study the changes of CR11 level in pregnancy and its correlation with the length of gestation . Secondly, we measure the level of plasma estradiol content in different pregnancy weeks ,to investigate the changes of estradiol and its relationship with plasma CR1-I .At last, by treating the cultured human trophoblast cells with CR11 and its inhibitor we study the effects of CR11 on estradiol release by trophoblast cells. Results: 1 .CRH concentration in premature and prolonged maternal plasma is respectively higher and lower ,as compared to normal pregnancy woman.Matemal plasma CR11 at 28 weeks correlates with the length of gestation(r= -O.827,p < 0.05). 2. plasma estradiol level in preterm and prolonged maternal plasma is respectively higherand lower than controls',p1asma eStradiol concentration has positive correlation withCRH level(re0'846,P < 0. 05). 3.CRH can increase estradiol release by cultUredtrOphblast cells as CRH chbitor reduce it.Conclu8ion:Can is the imPortat trigger factOr of hUman parturihon,one of itsmechanism is that CRH can stimulate estradiol release in trophblast cells by pararineand ProW parturition. |
