| Objective:To investigate the injunious effects of high concentration of superoxide caused by CTX and the protective effects of Superoxide Dismutase(SOD) and Granulocyte Colony-Stimulating Factor (G-CSF) on the bone marrow cells and the peripheral blood cells of mice. Methods:Mice were administered introperitoneally normal saline,low concentration (37.25 mg/kg), middle concentration (56 .25mg/kg) and high concentration (75.00mg/kg) CTX respectively. After zero,two,four and six days,bone marrow and peripheral blood cells were counted.The contents of 07 generated by the cells were measured by Cyt.c reduction,namely adding bone marrow and peripheral blood 1 x ~ 0~ cells to 2m1 of RPMI- 1640 culture fluid (without phenol red, and containing Cyt.c l6Oum). The cells were cultured at 370C.Take out the supematant and the absorbance was measured at SSOrim. Put mice bone marrow cells and peripheral blood cells (lxl0~) cells into 2m1 of RPMI-1640 culture fluid (containing 3}J~ TdR), and the cells incubated at 370C. The cells were harvested, and their radiactivities (DPM) were measured. The mice that were administered introperitoneally high concentration CTX were administered introperitonelly normal saline~, SOD and G-CSF, then repeat the count of cells, 02 generated and DPM. Results: CTX significantly increased the quantity ofas compared with that of control group. However, CTX significantly decreased the count of bone manow~ peripheral blood cells and DPM. After adding SOD to clean Oy-generated, the count of bone marrow cells were 5.568 times as compared with that of without SOD group, and Ofgenerated were 0.361 times, DPM were 2.829 times. The count of peripheral blood cells were 4.967 times as compared with that of without SOD group, and 01-generated were 0.481 times, DPM were 1.777 times. After adding G-CSF, the count of bone marrow cells were 9.177 times asII2compared with that of without G-CSF group, and Oy-generated were0.291 times, DPM were 3.37 1 times. The count of peripheral blood cells were 9.298 times as compared with that of without G-CSF group, and 07-generated were 0.33 7 times, DPM were 2.060 times. Conclusion: High concentration of Superoxide on caused by CTX was the one of mechanisms that led to damage action to mice bone marrow and peripheral blood cells.
