Accodingly,we undertook studies to examine the expression of CC chemokine in RA tissues and its role in inflammatory cell attacking rheumatoid synovium with the methods of flow cytometry and iminunofluorescence double-staining techniques. RA tissues obtained from 8 RA patients and 3 AS patients were detected. The expression of macrophage inflammatory protein (MIP fB )and RANTES was examined in CD3 CD4-. CD2ON CD68 cells in RA peripheral blood (PB) .. synovial fluid (SF) and synovial tissues (ST). Our results identified elevated levels of both MIP and RANTES expression in RA ST and SF T cells and macrophage (Mac). Moreover, the expression of MIP f3 in cultured Mae and T cells of ST and SF was much more increased in response to IFN .Whereas, the expression of RANTES in PB decreased after the stimulation of IFN.Histological examination of affected rheumatoid joints revealed extensive MIP1 f3 expression in sites of lymphocyte infiltration and cell proliferation, namely the synovial lining and sublining layers. MIP f3 was expressed in the plasma of CDY. CD4. CD2O CD68 lls as well as on the surface of the microvasculature endotheliocytes in RA ST. These data suggest that MIP P and RANTES which overexpressed in synovium line and its subline where synoviocytes proliferation and lymphocytes infiltration were significien, may play a key role in the selective chemoattractant and activation on leucocyte in RA.
