Objective To establish the method of constructing the bioartificial renal tubule assist device(RAD) by porcine proximal renal tubule cell line(LLC- PK1) and investigate the feasibility of creating the RAD in vitro. Methods The cultivated cells of LLC-PK1 were used .After the cells were seeded into the lumen of the hemofilter(7FH66),the filter was kept on incubating under the culture condition for a week.Then the filtration process was performed in vitro to detest the function of the RAD.The reabsorptive rate of sodium,creatinine and glucose at different time point were measured.The effects of oubaine and phlorizin on the reabsorptive i-ate of sodium and glucose were also detected.The morphology of the cells were observed by the electronic microscope. Results The creatinine reabsorptive rate of RAD group compared with negative one were significantly decreased (6.3 l.390/o vs 89.9 5.65%, P0.05).Afler oubaine and phlorizin were added into the substitute fluids, the reabsorptive rate of sodium and glucose significantly decreased as compared with control group( Nat: 11.3 2.07% vs 88.5 9.3 1%; Glu :7.2 0.58% vs 90.7 3.62%, P<0.01). Then the fresh substitute fluids without oubaine and phlorizin were used again.The reabsorptive rate of sodium and glucose could recovei(Na:83.6 7.73% vs 85.9 4.8 1%; Glu :83.9 .33% vs 90.2 9.07%, P >0.05). Under the electronic microscope the cells adhere to the lumen of the hollow fiber could be found. Conclusion RAD can be established by using LLC-PK1 cellline and detected the selective reabsorptive function of renal tubule in vitro.
