Objective:The expression and clinical significance of VEGF, KDR, MMP-9 and protein in invasive cervical carcinoma were investigated.Method:The expression of VEGF, KDR, MMP-9 and CD]05 protein, micro vascular density(MVD) labeled by CDs4 protein and proliferation index of cancer cells labeled by Ki-67 protein in 75 cases of invasive carcinoma of cervix(ICC), 18 cases of cervical intraepithelial neoplasia(CIN) and 15 cases of normal cervical epithelium remote from tumor(NCE) were detected by immunohistochemistry SP method, and the relationship between the expression of VEGF, KDR, MMP-9, CD!05 in ICC and MVD, proliferation of cancer cells, cancer invasion, metastasis and many factors relating clinical pathology of invasive cervical carcinoma was analyzed statistically.Result:The staining of VEGF, KDR, MMP-9 and CDios was mainly confined to the cellular membrane and / or cytoplasm in tumor cells, whereas that of Ki-67 was mainly confined to the nucleus, and CD34 was mainly confined to the vascular epithelial cells in tumor stroma. It was showed that the rate of positive expression of them increased remarkably in the order of NCE, CIN and ICC(P<0.05). hi CIN and ICC, the MVD was significantly higher when overexpression of VEGF, KDR,MMP-9 and CDios was taken place(P<0.05).In ICC, the MVD was positively correlated with the expression of VEGF, KDR, MMP-9 and CDios protein(P<0.05). The firstly strongest relationship between them and MVD was VEGF(r=0.602); secondly, CD105(r=0.499); thirdly, KDR(r=0.331); finally, MMP-9(r=0.287). The MVD was in higher level in patients with all positive expression of VEGF, KDR, MMP-9 and CD^s protein than that in those with other condition(P<0.01). The univariate analysis indicated that all the four factors had a significant effect on the MVD, and the main effect of them on the MVD was 50.8%. In addition, the expression of VEGF was correlated with KDR, MMP-9 and CDio5(P<0.05), and there was remarkable interaction between the expression of VEGF and CD,05(P<0.01). The expression of VEGF in ICC may play a critical role in tumor angiogenesis, and the expression of the others may be dependently cooperative action.There was relationship between the expression of VEGF, KDR in ICC and histopathological grading, pelvic lymphonode metastasis, intravascular involvement or expression of Ki-67(P<0.05), but no relationship between the expression of them and age, the international federation of gynecology and obsterics staging(FIGO staging), histopathological types, stroma involement, gland involement and tumor size(P>0.05). The rate of positive expression of them in the cases with grading III, pelvic lymphonode metastasis, intravascular involvement or overexpression of Ki-67 was greatly higher than that in those without them (P<0.05).The positive staining of MMP-9 was correlated with staging, grading, pelvic lymphonode metastasis, stroma involement, intravascular involvement or expression of Ki-67(P<0.05), but not correlated with age, types, gland involement and tumor size(P>0.05). The rate of positive staining of it in the cases with staging II, gradinglll, pelvic lymphonode metastasis, deep stroma involement, intravascular involvement or overexpression of Ki-67 was remarkably higher than that in those without them (P<0.05).There was relationship between the staining reaction of CD^s and stroma involement, intravascular involement or Ki-67 expression(P<0.05), but no4relationship between the staining reaction of it and age, staging, grading, types, pelvic lymphonode metastasis, gland involement and tumor size(P>0.05). The rate of the staining reaction of it in the cases with deep stroma involement or overexpression of Ki-67 was significantly higher than that in those without them(P<0.05). The percentage of the strongly expression of CDios in patients with intravascular involement was also higher than that in those without it(P<0.05).Conclusion:The result in this study indicated that the expressions of VEGF and its receptor KDR, MMP-9 and CDios in ICC were...
