Regulation On Genital Hormones And Receptors With Nourishiing-Kendey Herbs On Anovulatory Rats | | Posted on:2003-08-15 | Degree:Master | Type:Thesis | | Country:China | Candidate:Y C Duan | Full Text:PDF | | GTID:2144360065450242 | Subject:Traditional Chinese Medicine | | Abstract/Summary: | PDF Full Text Request | | Objective: To explore the mechanism, link and locus of Bushentiaojing recipe (consisting of Radix Rehmanniae Praeparata, Rhizoma Dioscoreae, Rhizoma Curculiginis, Epimedium, Human Placenta and others) in treatment of anovulatory disease, and provide theoretical evidence for clinical use of nourishiing-kendey herbs in inducing ovulation.Method: Sixty SD female 9 days old rats born in same batch were selected, and were randomly divided into four groups. One group which was injected neutrality tea-oil 0.05 ml each rat in nap by subcutaneous injection was to be normal group( group D). The other three groups( group A, B and C)were injected teststerone 0.05 ml each rat in nap by subcutaneous injection to make annovulatory rat models. Vaginal cells smear, which lasted two sex cycles (5 days every cycle ), was sequentially done since the seventieth day. The annovulatory rat models have successfully made when vagina epithelia was lasting keratinocyte. Group A was given nourishiing-kendey and regulating menstruation herbs 1 ml/lOOg weight ( It was 20 times of clinical use ) one time aday since the eightieth day. Group B was given wujibaifengkoufuye 0.5 ml/100g weight ( It was 20 times of clinical use) after diluted one time with distilled water. Group C and D were given distilled water 1 ml/100g weight one time a day since the eightieth day. Vaginal cells smear, which lasted two sex cycles was done after seven weeks. The rats were killed to take samples after vaginal cells changed. (1) The serum was detected by radio immunassay. They were Estradiol(E2), progesterone(P), follicle stimulating hormone(FSH), luteinizing hormone(LH) and prolactin(PRL). (2) Left ovary and left uterus were weighed. (3) Right ovary and right uterus were put into 4% formaldehydum polymerisatum to be fixed and embeded, and then used in immunohistochemistry and HE staining. So as to observe ovary and uterus morphological changes and their expressive intensity of estrogen receptor (ER) and progestogen receptor(PR).Results: FSH in group A was significantly higher than that of group C (p<0.01). There were no significant difference between group B and group C. LH in group A was higher than that in group C (p<0.05). There were no significant difference between group B and group C. PRL in group C was higher than that in group A and B (between group A and group C p<0.01, between group B and group C (p<0.05). Compared with group D, E2 in group C decreased (p<0.05). There were no significant differencebetween group A and group C , group B and group C. P in group D was higher than that in other three groups, the others showed no significant difference. The changes of ovary and uterus morphology were as follows: between group A, D and group B, C, follicles were at different levels; stratum granulosa were thick, and interstitial glands in ovary were hyperplasia, and endometrium was thick, the total of glands were improved. Ovary immunohistochemistry of ER and PR: The average gray degree of ER in group A granulosa was lower than that of group B and C (p<0.01). There were no significant difference between group B and group C. The average gray degree of ER in group A and group B luteum was lower than that of group C (p<0.05). The average gray degree of PR in group A granulosa was lower than that of group B and C (p<0.05). There were no significant difference between group B and group C. The average gray degree of PR in group A luteum was lower than that of group B and C (p<0.01). There were no significant difference between group B and group C. Uterus immunohistochemistry of ER and PR: The average gray degree of ER in group A gland and stroma were lower than those of the other three groups (p<0.01). The average gray degree of ER in group A endometrium and myometrium were lower than those of group B and C (p<0.05). The average gray degree of PR in group A endometrium, gland and stroma were lower than those of group C(p<0.05). The average gray degree of PR in group Amyometrium was lower than those of group C and D (p<0.05).
