Objective:Connective tissue growth factor(CTGF) has been implicated in the process of renal fibrosis by stimulating renal cell proliferation and accumulation of extracellular matrix. The effect might be influenced by high glucose angiotensin II (Ang II) or thus by angiotensin II receptor antagonist. In this study,we observe the effect of high glucose Ang II and Lorsatan on the expression of CTGFmRNA in cultured mesangial cells(MCs).Methods:Mesangial cells of SD rats were isolated and cultured. High glucose(30mmol/L) and Ang II (10-9 10-7- 10-5mol/L) were added to the medium for 72 hours to observe the influence on CTGFmRNA expression. Lorsatan of10-5mol/L and Ang â…¡ of 10-5mol/L were added to the medium to observe the effectsof Lorsatan on CTGFmRNA expression stimulated by Ang â…¡. The expressions ofCTGFmRNA were detected by means of reverse transcriptase polymerase chainreaction (RT-PCR).Results: RT-PCR showed that high glucose and Ang â…¡ up-regulated the expressionof CTGFmRNA,and Ang â…¡ stimulated the expression in a dose-depended manner.Expression of CTGFmRNA induced by Ang â…¡ was partially suppressed by Lorsatanat the concentration of 10-5mol/L(P<0.05).Conclusion: High glucose and Ang â…¡ can enhence the expression of CTGFmRNAand thus is involved in the process of renal fibrosis. Lorsatan can have a partialfibrogenesis-inhibiting effect,and thus implication in the treatment of renal fibrosis.
