| Objective To study the effect of Dracorhodin on high glucose-induced connective tissue growth factor (CTGF) formation in human mesangial cells (HMC), and its mechanism of prevention and treatment on renal fibrosis in diabetic nephropathy (DN) .Methods The HMC were divided into normal Group (NG group, 5.5 mmol / L D-glucose), high glucose (HG Group 25 mmol / L D-glucose), each group was located with Dracorhodin (7.5 umol / L) as comparison, and was examined at 24h and 48h .The expression of CTGF mRNA was assessed by semi quantatiue RT-PCR, and the expression of CTGF protein was assessed by Western blot.Results Compared to NG group, the expression of CTGF mRNA and protein were increased in HG group after stimulating 24 hours and 48 hours (P <0.01). Compared to HG group, the expression of CTGF mRNA and protein were reduced in HG group added with Dracorhodin after stimulating 24 hours and 48 hours (P <0.01, P <0.01).Conclusion Low-sugar environment, HMC can not be induced by excessive expression of CTGF, Dracorhodin in perchlorate intervention, the corresponding expression of no significant difference. HMC high glucose can increase the expression of CTGF, high-sugar up the difference was significant. Dracorhodin in perchlorate intervention after 24 h, high-sugar group in Canada's Blood CTGF mRNA and protein expression are down, had a significant difference. Dracorhodin in perchlorate 48 h after the intervention, high-sugar group in Canada's Blood CTGF mRNA and protein expression are lower, the difference was significant. These results show that: HMC can be induced by high glucose in the over-expression of CTGF, Xuejie after the intervention of perchlorate can inhibit CTGF, and a time-dependent inhibition of the expression of CTGF, DN control of renal fibrosis. This new way of treatment, at home and abroad has not been reported. |
