OBJECTIVE To study the effects of ethyl acetate extract of Sorbaria sorbifolia on acute and chronic hepatic injury induced by carbon tetrachloride (CCl4) and on precarcinogenesis induced by diethylnitrosamine (DEN). METHOD Sorbaria sorbifolia was administrate ig. to rats with CCl4-induced acute hepatic injury, hepatic fibrosis and DEN-induced precarcinogenesis, respectively then the biochemical indices were investigated. The activities of glutamic pyruvate transaminase (GPT), glutamic oxaloacetic transaminase (GOT), superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), nitric oxide synthase (NOS), and the concents of malondialdehyde (MDA), glutathine (GSH) and nitric oxide (NO) were investigated by colorimetric method; the levels of tumor necrosis factor (TNF-α) and hyaluronic acid (HA) were studied by radioimmunoassay. RESULTS Sorbaria sorbifolia reduced the elevated levels of serum GPT and GOT, increased the SOD, GSH-PX levels and decreased MDA level in serum, liver homogenate and liver mitochondria of rats with acute hepatic injury induced by CCl4; increased SOD, GSH-PX activities, elevated TNF-α, NO levels, decreased MDA,NO level in the hepatic fibrosis model; also increased the serum SOD, GSH-PX activities, decreased serum NOS and NO, MDA levels of rats in early stage of hepatocarcinogenesis induced by DEN. CONCLUSION Sorbaria sorbifolia extract has the function of protecting the acute and chronic hepatic injury induced by CCl4, and inhibiting hepatic precarcinogenesis induced by DEN.
