The Effect And The Mechanism Of Kuijieling On Experimental Ulcer Colitis In Rats

BACKGROUND AND AIM ulcer Colitis (u. c) is onekind of inflammatory bowel disease (IBD). Recently , It is the common disease in digest tract. The incidince of U. C is high in Europe and U. S and it is more and more higher in China. In modern medicine, Salazpsulfa pyridine (SASP) was first choosed, and the hormone and immunosuppressant also were used if the condition of the disease is serious. Although the drugs had their advantage, they had the serious side-effect . The study showed that the Chinese herbs, especial the recipe can get good effect and reduce the side-effect.Kuijieling (KJL) is composed of six kinds of herbs .which was created by professor Laoshaoxian, a famous TCM doctor in Guangdong province. The recipe can expel the heat and dampness and promote the circulation of the blood .The effect of kuijieling has been valuated in clinic. In order to valuate the effect of recipe, we investigated the effect of KJL on treating experimental U. C in rats , inhibiting inflammory, easing pain, antagonisting diarrea and regulating gastrointestinal motility .Methods ?U.C models were induced by Freunds , dinitrochlorobenzene(DNCB) and acetic acid, followed by the treatment with administration of Kuijieling orally in the dosages of 9.18g/kg, 4. 59g/kg and 2. 30g/kg. Then the histopathological changes including the ulcer, erosion and the soakage of lymphocyte were measured under optical microscope. (D Radio-immunity method was used in measuring the content of IL-6 and TNF-a . ㏕he animal models of granulation induced by cotton ball and swelling of rat foot induced by egg were used for the experiments of anti~inflammory . The animalmodels of pain response induced by heat and acetic acid also used for the experiments of easing pain the animal models of diarrhea induced by cantor oil and Senna Folia were used for the experiments of treating diarrhea. ㏕he charcoal ink-induced intestinal propellant movement in mice in vivo and colon contraction in rabbit were studied.Results (1) The pathologic changes of colon mucosa were greatlyimproved, and inflammative degree of mucosa and the content of cytokines (IL-6 and TNF-a) in treated group were significantly decreased compared to untreated group. (2)Kuijieling inhibited the degree of swelling of rat foot , decreased the weight of granulation induced by cotton ball and reduced the pain response . (3)The results showed that Kuejieling decrease the quantity of diarrea .(4) Kuijieling could inhibit the gastrointestinal propellant rate of charcoal ink in mice and Ach, BaCl2-induced spasmodic contraction of isolated rabbit intestine.Conclusion KJL has better effect for treating experimental U. C. Themechanism may be relate to anti-inflammory .easing pain, inhibiting inflammative factors(IL-6 and TNF-a ) and regulating gastrointestinal motility.
