Background and Objective Solid tumors require neovascularization for growth and metastasis.Thymidine phosphorylase(TP)/Platelet-derived endothelial cell growth factor(PD-ECGF) is a newly angiogenesis factor and its expression is high in various human malignant tumours.TP/PD-ECGF is a prognostic factor of colorectal or gastric carcinoma.lt is highly related to microvessel density(MVD) of gastric carcinoma. The expression level of TP/PD-ECGF play an important role in the action of 5-fluorouracil(5-FU) and its prodrugs.There are colse relations between the process,character,density of microvessel and the ability of tumou invasion,metastasis.Among all kinds of vascular endothelial cell markers,the specificity of CD34 is highestThere are few studies about MVD in gastric carcinoma,especially by use of CD34 monocolon antibody labelling vascular endothelial cell.The aim of this study is to assess the expressions of TP/PD-ECGF and CD34 in gastriccarcinoma,to investigate the correlations between TP/PD-ECGF,MVDand clinicopathological factors and the relationship between MVD andTP/PD-ECGF in gastric carcinoma.This study will provide helps forchemotherapy and antiangiogenic therapy.Methods The expressions of TP/PD-ECGF and CD34 antigen in 52cases of gastric carcinoma tissues, 10 cases of tumour borderline tissuesand 16 cases of normal gastric tissues were examined by use of S-Pimmunohistochemistry.To calculate the number of microvessels labelledby anti-CD34 monoclonal antibody.All samples had been proved bymeans of pathological diagnosis.Results l.In gastric carcinoma tissues,the positive rate of theTP/PD-ECGF expression (80.77%,42/52) is significantly higer than thatof normal gastric tissues (6.25%,l/16)and tumour borderlinetissues(30.0%,3/10)(P<0.005).l.There are not corelations between the expression of TP/PD-ECGF andthe age,gender of patients, the size,the extent of differentiation,the depthof invasion,lymph node or distant organs metastasis and TNM stage oftumor(P>0.05).3.Staining of CD34 is easy to identify because of its highspecificity,sensitivity and clear background.4.The mean of MVD in 52 cases of gastric carcinoma tissues is28.00,from 4.00 to 51.17. MVD of gastric carcinoma tissues(28.00+l 1 .82)is significantly higher than that of normal gastric tissues(12.85+10.30,P<0.01)and tumor borderline tissues(18.37+12.35yP<0.05).5. MVD of gastric carcinoma is related to the size of tumor. MVD withthe diameter exceed or equal to 5.0 centimeter(33.02+12.76) is significantly higher than that of with the diameter less than 5.0 centimeter(25 .34+1 0.53,P<0.05).6. MVD of gastric carcinoma is related to the depth of tumor invasion. MVD of gastric carcinoma tissues with muscle layer neoplastic involvement or subserosal neoplastic involvement(29.59+11.20),serosal neoplastic involvement (28.17+11.86),serosal involvement with directly infiltrating other organs beyond serosa(31.22+10.67) are all significantly higher than that of mucosal neoplastic involvement or submucosal neoplastic involvement( 1 2.03+4.55P<0.0 1 ).7. MVD of gastric carcinoma is related to distant organs metastasis of tumor. MVD of gastric carcinoma with distant organs metastasis(37.09+10.70) is significantly higher than that of without distant organs metastasis(26.09+l 1 .24P<0.05).8. MVD of gastric carcinoma is related to the TNM stage of tumor. Compared with I of TNM stage(23.17+11.50)and II of TNM stage(23.13+11.03),IV of TNM stage (36.42+10.31) is significantly higher(P<0.05). There is a trend of MVD gradually increase with increase of TNM stage.9. There are not corelations between MVD of gastric carcinoma and the age,gender of patients, the extent of differentiation,lymph node metastasis oftumor(P>0.05).10.
