In this article we study the relation of UPA, , CD34 to invasion and metastasis of gastric cancer and their relationship.Materials and Methods54 gastric cancer specimens were selected from paraffin wax embedded specimens with full clinicopathological data. They are divided into different groups according to histological differentiation . 15 cases of normal gastric muco-sa were studied as control. Immunohistochemical staining for UPA, CD34 was performed with a standard avidin - biotin - peroxidase complex detection kit. Statistical significance was calculated by t - test, x~2 test, relative analysis. The level of significance was set at P < 0.05.ResultUPA is high expression in gastric cancer and with the decrease of differentiation, lymphatic metastasis,long distance metastasis, increase of clinical stage, the expression is increase( P < 0.05).CD34 is high expression in gastric cancer and with the decrease of differentiation, long distance metastasis,increase of clinical stage, the expression is in-crease( P < 0.05 ).In gastric cancer the relativity of UPA and CD34 expression is positive, r = 0.411,P < 0.01.Discussion1. The test approve that in gastric cancer with the increase of malignancy the UPA expression increase. The result suggest that through some mechanism the ECM is hydrolysis by the increased UPA, and this inaugurate a invasive pathway. The finding is that UPA expression focus on the interface of cancer tissue and normal tissue. Cancer cell and normal cell are both high expression. So we can speculate that UPA high expression is the result of inducement and promotion of cancer cell and host cell.2. The test approve that the CD34 expression increase with the increase of malignancy in gastric cancer. The result suggest that MVD increase can obviously promote the invasion and metastasis of gastric cancer. We can infer that the new microvessel is induced by gastric cancer cell. CD34 is an antigen that has close relativity with new, immature microvessel . In new, immature microvessel CD34 express highly, and promote microvessel grow. New microvessel structure is not perfect, they can not only increase the invasion area, but also promote cancer cell fall into blood and cause blood metastasis. CD34 has no obvious relativity with cancer lymphatic metastasis, this may because CD34 can not give effective mark to lymphatic endothelium. According to the result we can believe that CD34 can become a new mark to diagnose, evaluate and examine gastric cancer.3. Result analysis show that the relativity of CD34 and UPA expression is positive, r = 0.411, P < 0.01, this suggests CD34 and UPA have coordinate action and promote each other. UPA inaugurate more space for new vessel creation, therefore CD34 can promote vessel creation.Conclusion1. UPA, CD34 have very important promoting effect during gastric cancer occurring, developing, invasion and metastasis.2. UPA and CD34 have coordinate action and promote each other during gastric cancer occurring, developing, invasion and metastasis.3. UPA and CD34 all can be regarded as indexes for gastric cancer diagnosis and prognosis evaluation.
