Liu Chuntao Associated Prof. Postgraduate: Wang WenzhiObjectives to study expression of CC16mRNA in induced sputum of asthmatics during exacerbation and stable stage, and compared it with IL-5 in induced sputum, to explore the role of CC16 and IL-5 in asthma. Methods Randomly selected 32 asthmatic patients who excluded the other pulmonary diseases and have not taken steroids for at least 1 months. Among them 8 subjects were in asthmatic attack, the others were in stable stage, selected 10 healthy subjects as control. Measured their lung function FEV1/FVC and induced sputum with 4% saline 20ml. dissolve the sputum with 10% DDT and extract the total RNA from the sputum cells, design CC16 primer and select ? -actin as control and ampllificate with RT-PCR. Measured IL-5 in sputum supernate.Results (1) total cell number and eosinophil number were higher in asthmatics in exacerbation than the other group. (2)IL-5 level in asthmatics in exacerbation was higher than that of the stable stage and control group, and their FEV1/FVC% was lower than that of the latters. CC16 level in asthmatic groups were lower than that of the control, especial lowest in exacerbation group. (3)according the course of asthma, divided the stable condition group into two groups and found that patients with a long duration of disease OlOyears) have lower CC16 level compared with the patients with a short duration ofdisease(<10year). there were no differences in IL-5 and FEV1/FVC% between two groups. (4)correlate analysis reveal that the number of eosinophil was correlated with CC16(r=-0.321) (p<0.05) and IL-5(r=0.861) (p<0.05) ; IL-5 was correlated with CC16(r=-0.461) (p<0.05) and FEV1/FVC%( r=-0. 98) (p<0. 05) ; CC16 was correlated with FEV1/FVC%( r=0. 475) (p<0.05).Conclusions induced sputum technique is a safe and effective method as reseaching airway inflammation. IL-5 and eosinophil have important role in asthmatic pathogenesis and propagate the asthmatic attacks. CC16 as an endogenous protective factor , it has been shown to have immunomodulatory and antiinflammation activity and play a role in controlling airway inflammation.
