Objective: To study the correlation between the airway remodeling and expressing of TGF-β1,IL-5,IL-13 in induced sputum of children with actue asthma.Method: 30 cases hospitalized children with acute asthma,15 cases healthy children were served as the control group.Ultrasonically nebulized 5% hypertonic saline was used to induce sputum.The IL-5,IL-13 level in induced sputum was determined using ELISA.immunocytochemistry assay was used to determine TGF-β1 expression of inflammtory cells.The forced expiratory ratio(FEV1) and the eosinophils(EOS) counting in the sputum were measured. The percentage of airway wall area to total airway cross sectional area(WA%) and wall thickness to airway diameter ratio (T/D) were calculated on high resolution computed tomography.Result: 1. The EOS count and expression of IL-5 and IL-13 were higher in asthma group than in control group,P﹤0.01 respectively. 2. 27 cases showed positive expression of TGF-β1 in inflammatory cells in induced sputum of asthma group,there is no positive expression of TGF-β1 in control group,furthermore,the ratio of TGF-β1 positive expression showed linear correlation with EOS count and WA%( r=0.655 P﹤0.01,r=0.409 P﹤0.05) respectively negative correlation with FEV1%pre(r=-0.684 P﹤0.01). 3.The IL-13 level in induced sputum of asthma group was positively correlated with EOS counting and WA% (r=0.407 P﹤0.05,r=0.438 P﹤0.05)and significantly negatively correlated with FEV1%pr(er=-0.466 P﹤0.01).4. The IL-5 level in induced sputum of asthma group was positively correlated with EOS counting and expression of TGF-β1 in inflammatory cells(r=0.407 P﹤0.05,r=0.438 P﹤0.05,it had the similar change tendency with WA% but no significant correlation.5.Statistically negative association was observed between airway WA% and FEV1%pre of asthma group(r =-0.399 P﹤0.05). 6.T/D and WA% were different between asthma group and control group (P﹤0.05) respectively.Conclusion: 1. The over-expression of EOS count,IL-5,IL-13 and TGF-β1 of in induced sputum of asthma patients reflects the inflammation of asthma patient′s airway,which may provide evidences for diagnosing and treatment by further studing.2.IL-13 and TGF-β1 might play an important role in airway remodeling.3.The airway wall thickness and remodeling can be measured and assessed by HRCT . |
