Objective We hypothesis PPAR-γ and its ligand agonist?Rosigli-tazone has antiinflammation effect to airway inflammation of guinea pigs with acute asthma.Methods 31 guinea pigs were divided into 5 groups: matched control group , asthma group , DXM treatment group , Rosiglitazone treatment group , Rosiglitazone+1/2 DXM(l/2 DXM) treatment group and assaied the cell numbers and classification of bronchoalveolar lavage fluid(BALF) of all groups guinea pigs , observed the pathologic alteration of bronchi lung constitution of guinea pigs. Measure the expression of PPAR-y mRNA> COX-2 mRNA in bronchi lung constitution of guinea pigs by RT-PCR. Results1. The alternation of total cell, neutrophilic granulocyte and eosi-nophilic granulocyte in BALF of guinea pigs1) The numbers of total cell, neutrophilic granulocyte and eosinophilic granulocyte in BALF of asthma group were all higher than these of other groups except Rosiglitazone group(P<0.05, respectively);2) Total cell, neutrophilic granulocyte of Rosiglitazone group in BALF were higher than these of other groups except asthma group (PO.05, respectively), eosinophilic granulocyte was lower than that of asthma group significantly(P< 0.01), but it was not difference compared with other groups (P>0.05, respectively) ;3) Total cells > neutrophilic granulocyte and eosinophilic granulocyte in BALF of DXM group and 1/2 DXM group were not difference (P>0.05, respectively ) and were higher than these of control group, but were notIVdifference (P>0.05 , respectively) ; 2. Pathological alternation1) There were a lot of inflammatory cell infiltration in airway mucosa and submucosa in asthma group, mucosa hyperemia edema and airway wall thickening obviously, airway wall, mucosa and submucosa was thickening than these of other groups (P<0.05, respectively);2) The thickness of airway wall of Rosiglitazone group was lower than that of asthma group (P<0.05 ) , but was not difference compared with other groups (P>0.05 , respectively) and the thickness of mucosa and submucosa was higher than these of DXM group , 1/2 DXM group and control group (PO.05 , respectively);3 ) The thickness of airway wall, mucosa and submucosa of DXM group and 1/2 DXM group were thicking not obviously , and were not differ- ence compared with control group (P>0.05 , respectively) ;3. Expression of PPAR-y mRNA in bronchi- lung tissue of guinea pigs1) Expression of PPAR-y mRNA of Rosiglitazone group and asthma group were lower than that of DXM group and 1/2 DXM group and was not difference compared with control group (P>0.05, respectively) ;2) Expression of PPAR-y mRNA of DXM group and 1/2DXM group were higher than that of other groups (P<0.01 Respectively) .The expression of PPAR-y mRNA of 1/2DXM group was lower than that of DXM group (P<0.05) ;3 ) There was still little expression of PPAR-y mRNA in control group;4. Expression of of COX-2 mRNA in bronchi lung tissue of guinea pigs 1) Expression of COX-2 mRNA of asthma group was highestamong all groups (P<0.05, respectively) ;v2) Expression of COX-2 mRNA of Rosiglitazone group was higher than that of DXM group and 1/2 DXM group (PO.01, respectively) ;3 ) Expression of COX-2mRNA of DXM group and 1/2DXM group was not difference compared with each other (P>0.05 );4) There was not expression of COX-2mRNA in bronchi lung tissue of control group.Conclusion1. Rosiglitazone has the effect of relieve airway inflammation in guinea pigs with asthma, and the effect was more effective if used with DXM2. Rosiglitazone decrease eosinophilic granulocyte in airway of guinea pigs with asthma, and this maybe one of mechanisms of antiin-flamma- tion3. Rosiglitazone decrease the expression of COX-2 mRNA in bro-chi- lung tissue of guinea pigs with asthma4. Dexmethason decrease the expression of COX-2 mRNA in bronchi- lung tissue of guinea pigs with asthma;5. Rosiglitazone and DXM had synergistic action, and can reduce the dosage of DXM for half;6.
