| This paper is about the extraction process and quality assessment of Xuanfudaizhe decoction, which is composed of hedge glorybind flower, hematite, tangshen root, licorice root, ternate pinellia rhizome, common ginger rhizome, common jujube fruit.Britanin, one of the sesquitepene lactones, was isolated and identified from hedge glorybind flower. Standard britanin for quality control was prepared. A reversed-phase HPLC method was established to determine the content of britanin. Britanin was analyzed by a Spherisorb ODS column. The mobile phase was methanol-water (40:55, v/v) and detection wavelength was set at 210 nm.The results showed the calibration curve was linear in the range of 7.984-159.7 μg.ml-1 and correlation coefficient was 0.9998. The average recovery was 96.8% (RSD=1.9%). This method was simple, sensitive and reproducible and provided a quantitative basis for the quality assessment of hedge glorybind flower.Using water and ethanol of different concentrations as the extracting solvent, the effects on extracting efficiency with different extracting methodswere studied. The optimal process was studied by orthogonal design with content of the britanin, quercetin content and extractive yield from Xuanfu-daizhe decoction as indices. Six factors were selected in this experiment, including the water and ethanol consumption, duration of extraction and times of extraction. The result showed that the optimal extraction process was 95% ethanol consumpted 10 times the amount of material, refluxing for 2 times, 3 hour each time, and residue was water consumpted 8 times the amount of material, refluxing for 3 times, 1 hour each time.Britanin, quercetin in hedge glorybind flower of Xuanfudaizhe decoction were determined by RP-HPLC. They were analyzed by a Spherisorb ODS column The mobile phase was acetonitrile-water-tetra hydrofurant (22:78:1.5, v/v), methanol- acetonitrile- water-acetic acid glacial (15:15:70:1, v/v) and detection wavelength was set at 210, 254nm respectively. They had good linearity within the range of 4.168-62.52 μg.ml-1, 4.70-70.5 ug.ml-1 respectively. The average recoveries of britanin, quercetin were 100.5% (RSD=1.2%), 97.3%(RSD=1.5%) respectively. Five herbs of Xuanfudaizhe decoction were identified by TLC with the spots clear and reproducible. TLC and HPLC specification of Xuanfudaizhe decoction provided its method of quality assessment. |
