| Objective To evalute the nitric oxide (NO) and the nitric oxide synthase (NOS) in rabbits' lens .To invest the relationship between NO,NOS and cataract.To confirm the protect role of the taurine to rabbits'lens. Methods Rabbits' lenses cultured in vitro were divided into 3 groups: control group, Feton model (oxidative stress group), group treated with 1% taurine.Another group was the fresh rabbits' lens. After 24-hour incubation, the levels of the soluble proteins,calcium,NO and NOS were tested.The distribution of the cells which were positive in NADPH histochemistry stain in lens were examined under light microscopic.The pictures were analysed by ViDAS Medical Picture Analytical System. Results There were no significant differents between the fresh lenses and the cultured control group. After 24-hour incubation, the soluble proteins level was decreased and calcium, nitric oxide levels, nitric oxide synthase activities increased significantly in Feton model compared with control groups .The soluble proteins level was increased but others decreased in 1% taurine group compared with Feton modle.The NADPH positive reaction is royal blue.The statistical results of NADPH are the same as the biochemical results. Conclusions The NADPH positive cells include the lens epithelial cells and the lens cells (fibers).The NOlevel and NOS activity were increased during cataract formation.The NO can damage the lens with oxidation and make the lens opacity.The taurine , an antioxidant ,can inhibit the production of NO and protect lens against oxidation damage, preventing lens from cataract development.Postgraduate LIU Hongxia (Ophthalmology) Directed by Professor YAN Honglu... |
