The Investigation Of Hypertonic Citrate Adenine Soulution For The Preservation Of Human Pancreases Islet And The Compatible Relationship Between The Human Blood And Isolated Islets Of Langerhans

BACKGROUD To establish a new method of cutting humanpancreases from cadaver donors for islets transplantation ,and to discuss therelationship between activity of islets and the cold ischemia preservation timeafter hypertonic citrate adenine solution perfused . To investigate therelationship between the living of islet and HLA typing in the donor andrecipients ,when human islets were exposed to nonanticoagulated humanblood,it will induce a rapid consumption of platelets,neutrophils , monocytesand lymphocytes in the blood.when heparin were added to the blood ,theseevents were avoided.The consumption of platelets will how change with theHLA typing compatible , and the HLA typing of blood and islets influce toliving of islets.METHOD l,500-2,000ml of hypertonic citrate adenine solution was perfused into aorta, kidey- pancreases-duodenum -spleen were cut together, digestion of pancreases was done with collagenase P,then stained the islets and count the living islet. Put the islet which have know HLA typing into the blood whose HLA typing is compatible for it,and the blood whose HLA typing is incompatibility for it.Observe the variability of platelets,neutrophils monocytes and lymphocytes in the blood after cultivates 1 hour, the varieties of living islets after mixed-cultivation of 1 hour and 24 hours. RESULT Activity of islets and the cold ischemia preservation time is in a negative correlation ; Activity of islets and the ischemic index is in a positive correlation, when human islets were exposed to nonanticoagulated human blood,it will induce a rapid consumption of platelets,neutrophils , monocytes and lymphocytes in the blood.when heparin were added to the blood ,these events were avoided. After cultivates 24 hours , the quantity of living islets in HLA typing compatible groups is more than the groups which HLA typingis incompatibility.CONCLUSION the cold ischemia preservation time of the pancreases preserved in hypertonic citrate adenine solution is better not more than 5-6 hours, when human islets were exposed to human blood ,a series of inflammatory events were found to occur; HLA typing will conduce to enhance the quantity of living islets.
