The Espression Of BFGF,FⅧRAg,PCNA,ER,PR In Uterus Leiomyoma | | Posted on:2004-06-15 | Degree:Master | Type:Thesis | | Country:China | Candidate:J Xu | Full Text:PDF | | GTID:2144360092499629 | Subject:Obstetrics and gynecology | | Abstract/Summary: | PDF Full Text Request | | Objective : To investigate the expression of Basic fibroblast growth factor(bFGF),Factor Ⅷ related antigen(FⅧRAg),Proliferating cell nuclear antigen (PCNA),Estrogen receptor(ER) ,Profesterone receptor (PR) in uterus leiomyoma and the relationship between bFGF and Microvessel density(MVD),PCNA, ER, PR .Methods: To choose 30 specimens uterus leimyoma's tissue which was verified by pathology as benign uterus leimyoma and normal uterus muscle tissue of the patient . All specimens separated to 2 groups: (1) experimental group : uterus leiomyoma tissue . (2) control group : normal uterus tissue in patients of leiomyoma . All cases were detected by immunehistochemistry SP methods with polyclonal antibody against bFGF,FⅧRAg,PCNA,ER & PR .They were fixed quickly after being deprived, and were lied in wax . Then they were cut into slice . It appeared positive expression in cell liquid and nuclear.Results: (1) Expression of bFGF : positive expression of bFGF in leiomyoma cell liquid was pale brown and was observed as granulose matter . bFGF were highlight in leiomyoma, but it was low or negative expression in controlgroup. The expression of bFGF were 14.77±14,41%, 5.09±4.46% in experimental group, control group respectively . There were statistic difference between experimental group and control group. (2) The expression of FⅧRAg: FⅧRAg drained in microvessel is from pale brown to deep brown. In leiomyoma it was drained to pale brown. It was lower expressed in control group. MVD(which was calculated by measuring FⅧRAg) in leiomyoma, control group is 20.15±12.47%, 11.48±6.79% respectively . There was statistic difference between leiomyoma and control group. (3) The expression of PCNA: It is pale brown in cell nucleus. The expression of PCNA is higher in leiomyoma than in the control groups. They were 22.38±13.19%, 7.41±5.38% in two groups respectively. There were statistic difference between leiomyoma tissue and the control groups. (4) The expression of ER: The positive expression of ER is pale brown in cell nucleus. It was lower in control groups than experimental group. The positive rate is 0.95±0.75, 0.47±0.59 in the two groups respectively. There were statistic difference between leiomyoma tissue and the control groups. (5) The expression of PR: The positive expression of PR is pale brown in cell nucleus. It was lower in control groups than in experimental group. The positive rate is 1.02±0.38, 0.53±0.42 in the two group respectively . There were statistic difference betweenleiomyoma tissue and the control groups. (6) The correlation between bFGF and MVD,PCNA: It perform correlated analyze between MVD & bFGF in leiomyoma, we get the correlate coefficient r=0.89. They had closed relation. It perform correlated analyze between PCNA & bFGF in leiomyoma, we get the correlate coefficient r=0.79. They had closed relation. (7) The relationship between bFGF and ER,PR: It perform correlated analyze between ER & bFGF in leiomyoma, we get the correlate coefficient r=0.26. There were no relationship between bFGF and ER. It perform correlated analyze between PR & bFGF in leiomyoma, we get the correlate coefficient r=0.34. There were no relationship between bFGF and PR .Conclusion: The result of the study show: Protain bFGF strenthened in leiomyma tissue, which was related to MVD and PCNA. It was stated that bFGF as an important growth factor can improve the development of leiomyoma through fastening vascular formation and stimulating growth of leiomyoma cell.It is possible the initial step of the development of leiomyma and related closely to the development of leiomyoma.Through understanding the distribution of protein bFGF in leiomyoma, we can understand the regulation of growth of leiomyoma. Then we can put forward new treatment method through refrein the first step of leiomyoma .
