Rutoside (Ru) is a fiavonoid glycoside, which can be extracted from many herbs.The purpose of this stUdy is to investigate the prophylactic and theraPeutic effect of Ruon experimental acute pancreatitis (AP) and its possible mechansms. The main contentsare as fOllows:l. Prophylachc effect of Ru on experiInental acute pancreattis in ratsAP model was induced by retrograde injection of sodium deoxycholate intobiliopancreatic duCt or by ligating the biliopancreatic duct at the point of entry into theduodeum in rats. Ru (30, 60, l20mg.kg-') were admhaStered subcutaneously tWo hmesbefore the induction of AP ProPhylactic Ru sighficantly inhibited the increase of sennnamylase (AMS) and lipase (LPS) at 6h, l2h and 24h aller the induction of AP reducedthe pancreas index and ascites volume, driproved the survival rae, inhibated thepancreatic edem4 necrosis, inflammation and vacuoliZaion at 24h aller the induction.The acion indicated a dose-dependent manner2. TheraPeutc effect of Ru on exPerimental acute pancreatitis in ratsRu (30, 60, l20mg.kg') were administered subcutaneously ten minutes after theinduction of AP, then in a one hour interval fOr six hours. Ru 60, l20mg.kg-' markedlyinhibited the sean AMS and LPS elevation, reduced pancreas index and ascitesvolume, improved the pathologic histology of pancreas significantly3. Possible mechanisms of the antagonistic action of Ru on experimental APRu (30, 60, l20mg.kg') were adIninistered subcutaneously two times before theinduction of AP Thre doses of Ru could decrease the nitric oxide (NO) level andincrease the Ca2+ level in serm of AP rats. It also can enhance the suPeroxidedismutaSe (SOD) aCtivity and phOspholipase A2 (PLA2) content of pancreas tissue.MeanWhile, the Ca2+ concentration, and the malondialdehyde (MDA) production inpancreas tissue are much lower in the Ru treated grouP.The result of thes stUdy suggested that Ru possess certain protective effect on theexperiInental AP in rats. The mechAnsms might be due to strengthen endogenousanioxidaive system and the ability of scavenge oxygen-derived free radicals (OFR),block the tranSlocation of calciuxn from exbecellular to intracellular compartInents andattenUate the injury of Ca2+ overoad in acinus. Maybe, the antagoninic effect is alsorelated to the inhibition on PLA2 edivation.
