PURPOSE:To investigate the effect and mechanism of moschus and muscone on rat retinal neurons in vitro.METHODS:1 The retinal neuron suspension was made from 1-3 day SD rats. 7 groups of moschus and muscone solutions,the concentration of which was lg-L-1-0.015625g.L-1,and the sovent were added into the cell suspension respectively-After 24 hours,the OD of the living cells was tested by MTT assay. The toxicity of the sovent was to be evaluated. The top non-toxic concentration(rCo) of moschus and muscone on rat retinal neurons was to be calculated. 2 (1) 4 groups of moschus solutions,the concentration of which was 2:3:4:5,under its TCo,were added into the cell suspension. The period of culture was 2 and 4 days. Then,the OD of the living cells was tested by MTT assay in day 2 and day 4. The effect of moschus on rat retinal neurons was to be evaluated. In the same way,the cell suspension was treated by moschus associated with a certain quantity of m-NGF. and its effect was to be evaluated. (2) 4 groups of muscone solutions,the concentration of which was 2:3:4:5,under its TCo,were added into the cell suspension. In the same way as above,the cell suspension was treated by muscone and muscone associated with m-NGF,and their effects were to be evaluated. RESULTS:1 The sovent of moschus and muscone has no toxic effect on rat retinal neurons. The TCo of moschus on rat retinal neurons is 0.129g.L-1,and the one of muscone is 0.700g.L-1. 2 Moschus has no significant effect on the living of 2,4 day-cultured rat retinal neurons(P>0.05). When associated with m-NGF,moschus in some certain concentration such as 0.075g.L-1 and 0.100g.L-l,can promote the living of 2,4 day-cultured rat retinal neurons significantly(P<0.05). Muscone has no significant effect on the living of 2,4 day-cultured rat retinal neurons(P>0.05). When associated with m-NGF,muscone in some certain concentration such as 0.1875g.L-1 and 0.2500g.L-1,can promote the living of 2,4 day-cultured rat retinal neurons significantly(P<0.05),and in 0.2500g.L-1,it can promote the living of 4 day-cultured rat retinal neurons significantly(P<0.05).CONCLUSIONS:By enhancing the biological efficacy of nerve growth factor (NGF),moschus can promote the living of retinal neurons indirectly. Among all of moschus ingredients,muscone is one of the most effective,enhancing the biological effect of NGF,at least.
