Objective To investigate the DNA polymorphism of Penicillium marneffei (PM) and to supply a technique for PM classification. Methods Total DNA was extracted with benzyl chloride. Random Amplification Polymorphic DNA(RAPD) assay was used to study the finger-printing of 12 strains Penicillium marneffei. Results ①Of forty random primers used, four of them :P23:5'-AGTCAGCCAC -3',P103:5'-AGACGTCCAC -3',P105:5'-AGTCGTCCCC- 3',P120:5'-GGGAGACATC -3'gave good reaction. ②Most of the isolates DNA patterns had major bands with the same mobility. Bands of Amplified were Polymorphic. Similarities was 44.4%-97.3%.③12 strains were divided into 5 groups. Conclusion 12 strains PM (mycelium) DNA extracted from different area PsM patients were Polymorphic and they were divided into 5 groups by RAPD-PCR technique. And this study has guide significance for PM epidemiology, prevention and cure.
