OBJECTIVES To investigate the difference of the gene polymorphism as well as the pathogenic relations in Guangxi PM between the isolates obtained from Rhizomys pruinosus and those from penicilliosis marneffei(PSM)patients.METHODS Total DNA was extracted with benzyl chloride.Polymorphic microsatellite marker(PMM)analysis was used to study the gene polymorphism and the pathogenic relations of 43 isolates penicillium marneffei obtained from Guangxi Rhizomys pruinosus and 40 isolates obtained from PSM patients in Guangxi.RESULTS(1)The natural positive carrier rate of the 43 was 100%. (2)There were 86 different alleles from 83 samples about 8 pair of primers.The PMMâ… has the most,altogether 42 alleles;PMMâ…§has the least,is 8 alleles.It showed that the genetic diversity level of 8 microsatellite loci was high.(3)The genetic identity of Guangxi PM between the Rhizomys pruinosus and PSM patients is 0.8502,the heredity distance is 0.1623;The genetic identity of PM obtained from PSM patients in various districts of Guangxi is between 0.5516 and 0.8364;and from Rhizomys pruinosus in various districts of Guangxi is between 0.4503 and 0.6384;The genetic identity is between 0.2986 and 0.8364 in the PM from the different districts of Guangxi Rhizomys pruinosus and PSM patients,the heredity distance is between 0.1787 and 1.2086.(4)Simultaneously the genes were divided into 5 types according to UPGMA.CONCLUSIONS There is the high gene polymorphism in Guangxi PM between the isolates obtained from Rhizomys pruinosus and those from penicilliosis marneffei(PSM)patients.The pathogens cause of the hunman PSM probably closely related to the Guangxi Rhizomys pruinosus PM. |
