| The mechanism of aging and antiaging with ginseng saponin from stems and leaves was studied by the model mice of subacute aging that was induced by D-galactose.1. The model of aging was established by continuous nap subcutaneous injection of 120mg/Kg D-galactose on 2-month-old male Kunming mice for 40 days. The result showed that content of malondialdehyde (MDA) and activity of MAO-B in brain tissue of aging mice were increased significantly, activity of superoxide dismatase (SOD), nitricoxide synthase (NOS), ATPase and content of nitric oxide (NO) decreased remarkably. It could be found there were pyknosis of neuron, depolymerization of ribosome, accumulation of lipid droplets and lipofuscin, condensation of chromatin. For theglia cell, there were swelling of body, cavitation of cytoplasm, decrease of synapses, dissolution of filamentinaxon. It indicated that the quasi-aging mice induced by D-galactos provided a good biological model to study on antiaging.2. In order to observe the effect of GSSL on aging model mice, the mice received continuously GSSL 50mg/Kg d and 200mg/Kg d by pouring respectively them into stomach at the same time when injected D-galactose. It was found that proper dosage of GSSL could antagonize the effect of D-galactose on the mice. The results showed that content of MDA was obviously decreased and activity of MAO-B in brain tissue of the therapy mice were inhibited, and activity of SOD, ATPase and NOS and content of NO were increased remarkably. The ultrastructure of brain tissue of the mice ameliorated significantly under electron microscope. Large dosage of GSSL (200mg/kg) was not able to invert aging induced by D-galactose. It revealed that proper dosage of GSSL with the function of diplodirect regulation to bodies of animals could enhance action of antioxidation and energe metabolism in brains of them. In addition, it could adjust metabolism of neurotransmitter and reduce content of free radical of brain tissue, thereby the aging brain tissue could be improved and protected. |
