Panax quinquefolium L. is a perennial herb of Araliaceae, originally produced in north America. It is one of well known herbs worldwide. Panax quinquefolium is fistly listed in a work named as"A Supplement to Compendium of Materia Medica" and had been used for over several hundred years in our country. Numerous studies show that Panax quinquefolium L. has extensive biological activities and pharmacological effects in many ways including the cardiovascular, endocrine systems.American ginseng, i.e., roots of Panax quinquefolium, was mostly used as medicine in the past. In our country, lots of basic reseach of the stems and leaves of Panax quinquefolium was carried out since the 1990s. It was proved to have an equally important medicinal value. From its aerial part, domestic and foreign scholars also gained the main activity part-Panax quinquefolium saponins from steams and leaves (PQS) in which saponin content is richer than that in roots and saponin specie is different from roots. It provided important scientific evidences for the rational development and comprehensive utilization of the Panax quinquefolium L. resources.Through literature review and preliminary findings of the research group, we found that most of saponins isolated from PQS are common, such as ginsenoside Rd, ginsenoside Rb3, etc. But the trace saponins remains to be further studied, here is a systematic study on chemical constituents of the trace saponins of the stems and leaves of Panax quinquefolium L. was carried out by various chromatographies. It will lay the foundation for the establishment of more scientific quality standard and the construction of its chromatography fingerprint, and the development of new formulations.Nine compounds were isolated from the saponins of the stems and leaves of Panax quinquefolium by repeatedly applying to silica gel columns, ODS columns and prep-HPLC. On the basis of chemical methods and spectroscopic analysis including 1H-NMR,13C-NMR, their structures were elucidated as gypenoside LXXI (I), ginsenoside F2 (â…¡), gypenosideâ…¨(â…¢), vinaginsenoside Rg (IV), vinaginsenoside R9 (V), majoroside-F1 (â…¥); 3-O-β-D-glucopyranosyl-(1â†'2)-β-D-glucopyranosyl-20-O-β-D-arabinopyranosyl-(1â†'6)-β-D-glucopyranosyl-dammar-23-ene-3β,12β,20S ,25-tetraol (VII), gypenosideLXIX (VIII), ginsenosideRd2 (â…¨),. Compoundâ… andâ…§is first isolated from Panax quinquefolium L. Compoundâ…£,â…¤,â…¨were obtained from the steams and leaves of this plant for the first time. Compoundâ…¦is a natural product; The purities of compoundsâ…¡,â…¢,â…£,â…¤,â…¥,â…§andâ…¨were up to 98% respectively by analytical HPLC, and compoundsâ…¡,â…¢,â…£,â…§can be used as reference substances.
