| WBE" is a kind of functional Ca additive that was extracted from bovine thighbone. It has been patented (WO96/23805). The object of this paper is to determine the effect of WBE on Ca supply and detect its functional factors by animal feeding and cell cultivation.60 Kunming rats were randomly divided into three groups as control(CaCO3 additive), test 1 (common bone power additive) and test 2(WBE additive). And then feeding started.The results of Ca metabolism showed that the Ca absorption rate (86.16 10.24%) and the Ca reservation rate (94.12 7.14%) were of significant difference from the other two groups respectively.Length of thigh, crus, lumbar of rats was of marked difference in the three groups. And their average length of WBE was in turn 16.071mm, 17.939mm, 19.645mm.There lied remarkably difference of bone density in the different groups, especially that of WBE additive group rose to 1.24 0.17mg/mm3.The content of organic substance also significantly increased compared with the other groups (P<0.05) .The content of bone Ca, Zn, Mg of WBE additive group was respectively 230.16 6.5mg/g, 6.66 1.47mg/g, 290.48 41.76 g/g, and the Ca content was significantly higher than control.The biomechanical parameters of bone in the WBE additive group increased notably compared with the other two groups (P<0.05 P<0.01) .From the data of bone histomorpometric analysis, Trabecular bone in the WBE additive group increased and broadened, and bone cavity became narrow. On the contrary, Trabecular bone of the other two groups declined, and bone was thinner to break.After 48 h, the contents of free Ca and P in the fresh bone in turn were 3300 g/g, 4050 g/g, which remarkably smaller than the other groups.The contents of serum Ca, P AKP were respectively 7.78 0.17mg/dL, 6.37 0.19 mg/dL, 145.52 25.16 JU/dL, which were different from the others.In addition, BMP was extracted and purified from WBE. Following that, extract was done SDS-PAGE, whose results showed the three main protein strips, the molecular weight of which in turn was 14, 16, 30kDa. All above suggested WBEcontained the functional ?actor BMP.As far as cell cultivation, DNA content of WBE additive group increased to 12.3 g/dlsh, 10.2 g/FGM; the content of collagen rose to 85 g/dish, 82 g/FGM. The results were remarkably different from that of the other groups. The observation of FGM with optical and electronic microscope suggested that BMP could significantly facilitate proliferation of osteoblast and accelerate formation of bone.The result demonstrated that WBE was a first-rate Ca additive, which could improve Ca absorption and reservation, boost bone growth, amplify bone mechanics, prevent outbreak of osteoporosis. |
