Recent reports revealed that VEGF from tumor cells could inhibit the activation of NF-KB which depended on TNF-a in hematopoetic progenitor cell. So the NF-KB could not go into the nuclear and could not up-regulate the expression of HI?histone ,then affected the differentiation of HPC and the development,differentiation and function of dendritic cell in tumor bearing host. All these could induce abnormality of the host's immune system. In our studies we used immnohistochemistry methods and hybridization in situ to investigate the relation between VEGF and the invasion and metastasis of esophageal squamous cell carcinoma. We also studied the effects of VEGF on dendritic cell in this kind of tumor and its effects on the invasion and metastasis.Methods: The expression of VEGF MVD dendritic cells tissue inhibitor-2of matrix metallo-proteinase in forty-six specimens of esophageal squamous cell carcinoma were examined by immunohistochemistry staining (S-P method) and hybridization in situ ,using anti-VEGF monoclonal antibody,anti-CD34 monoclonal antibody,anti-S-100 polyclonal antibody,anti-TIMP-2 polyclonal antibody and labeled VEGF mRNA probes.Results:1. Twenty-seven out of forty-six specimens of esophageal squamous cell carcinoma (58.7%) showed positive expression of VEGF protein. There were remarkable difference of VEGF protein expression in tumor in different situation of invasion and metastasis. The positive rates of VEGF mRNA were 39.1%.2. The positive rates of TIMP-2 were 55.2%(24/46). There were significant difference between TIMP-2 expression and the grade and invasion of tumor. No association between VEGF and TIMP-2 were found in esophageal squamous cell carcinoma.3. The mean of MVD in tumor was 42.87?7.41/Area Unit. The MVD in VEGF positive expression tumor(50.71?2.16/Area Unit)were significant higher than those in VEGF negative expression tumor(1.74+17.93/Area Unit).There were also significant difference of MVD of tumor in different situation of invasion and metastasis.4. The mean density of dendritic cell in tumor was 19.83?3.91/HP. There were great difference of the density of dendritic cell in tumor of different invasion and metastasis situation. The density of dendritic cell of VEGF positive expression tumor(14.93?2.83/HP) were much lower than the tumor of negative expression(26.79?2.61/HP). There was a close negative relation between the expression of VEGF and the density of dendritic cell in esophageal squamous cell carcinoma(R=-0.462).Conclusion:1. The expression of VEGF protein in esophageal squamous cell carcinoma had relation with invasion and metastasis, but not differentiation of tumor cells. These might display that VEGF protein could promote the invasion and metastasis of esophageal squamous cell carcinoma.2. The expression of VEGF mRNA were lower than the expression of protein, these might display that the immunohistochemistry of VEGF protein were better than hybridization in situ when come to the clinical application.3. The expression of TIMP-2 protein in esophageal squamous cell carcinoma had relation with the invasion but not metastasis of tumor cell, which mean that the TIMP-2 protein might inhibit the invasion of esophageal squamous cell carcinoma.4. The expression of TIMP-2 had no relation with the expression of VEGF in esophageal squamous cell carcinoma.5. The MVD of esophageal squamous cell carcinoma had relation with the invasion, metastasis, expression of VEGF protein, but not the differentiation, which showed that MVD could use as a reference method to check the invasion, metastasis, prognosis of carcinoma.6.
