ObjectiveOral cancer in the world has become a universal problem. Oral canceraccounts for5%of systemic cancer incidence, are one of the ten most commonmalignancy in the world, of which,90%of oral cancer epithelial squamouscell carcinoma. According to incomplete statistics, every year about8,000patients died of the disease, including surgery, radiotherapy, chemotherapy,including a variety of methods are constantly improved, but patients with oralsquamous cell carcinoma (oral squamous carcinoma cell, OSCC) the five-yearsurvival rate of only50%, it is a serious threat to the life and health of thepeople.Domestic scholars have done a lot of research on squamous cellcarcinoma of PTEN and MMP-7and VEGF.1997American scholars on the10th human chromosome found a novel tumor suppressor gene, is ever found aspecific tumor suppressor gene-PTEN. Mutation in this gene inactivation isclosely related to the occurrence and development of a variety of humanmalignancies, plays a key role in the process of apoptosis, cell cycle and cellmigration. MMP-7as streptolysin A is the molecular weight of the smallestmember of the matrix metalloproteinase family, with a superior proteindissolving capacity and a broad substrate specificity. MMP-7is inactivated ina variety of tumor cells, can degrade type IV collagen, proteoglycans, casein,laminate, fibrositis protein. MMP-7through the lysis the FasL to prolong thesurvival of tumor cells, and could resist the adriamycin-induced Fas/FasL-mediated apoptosis of tumor cells. VEGF is a distinctive specificvascular endothelial mitogen, the role of the strong stimulation of endothelial cell proliferation. To enhance the permeability of blood vessels, promotetumor angiogenesis, tumor vascular endothelial proliferation and migrationplay a role in tumor proliferation and metastasis conditions. About PTEN andMMP-7and VEGF are mainly concentrated in giant cell tumor, and in OSCCrarely reported.To study the relationship between the expression of PTEN,MMP-7and VEGF in oral squamous cell carcinoma(OSCC)and the biologicalbehavior and prognosis of oral squamous cell carcinoma.Materials and methodsFormalin-fixed, paraffin-embedded blocks of40tissues with OSCCand40normal oral tissues were used for immunohistochemical staining ofPTEN, MMP-7andVEGF respectively. The expressions of PTEN, MMP-7,VEGF and other clinical fractures were analyzed with the metastasis andrecurrence of the OSCC1Experimental specimens are taken from the Department of Pathology,Liaoning pro-vincible People’s Hospital from January2005to December2011the OSCC cases of dental surgical resection, including25males,15females,aged41to85years, an aver-age of64years. All cases had chemotherapy,radiotherapy and immunotherapy.Pathan-logically wasdiagnosedwithOSCCparaffin-embedded specimens40cases,High,moderatelydifferentiated group of11patients,29cases of poorly differentiated group.Allspecimens were fixed in10%neutral formalin solution, embedded in paraffinand paraffin-embedded specimens of40cases in the same period in surgicalresection of adjacent normal oral mucosa as a control group.2Experimental MethodsTest samples are serially sectioned, thickness:4μm, respectively, forHE staining and PTEN, MMP-7and VEGF immunohistochemical staining. Allstaining are located positive control (with tonsil tissue the PTEN positivecontrol; placental tissue MMP-7positive control; colon cancer tissueVEGFexplicit control; with PBS instead of primary antibody as negativecontrol).Results1、PTEN expression in OSCC was45%(18/40),85%(34/40) in normaloral tissue, the difference was statistically significant (P <0.01); MMP-7and VEGF in OSCCThe positive expression rate of75%(30/40),70%(28/40) innormal oral tissues were22.5%(9/40),25%(19/40), the difference wasstatistically significant (P <0.05).2、 PTEN was negatively correlated (P <0.05); MMP-7and VEGFexpression was positively correlated with pathological grade (P <0.05).3MMP-7and VEGF expression in OSCC were positively correlated (P <0.05). PTENexpression in OSCC was negatively correlated with MMP-7and VEGF(P <0.05).ConclusionPTEN down regulation/deletion with OSCC were closely related to theoccurrence and development; MMP-7and VEGF up regulation was closelyrelated to OSCC.Measurement of the expression of PTEN,MMP-7and VEGFmay have clinical significance in understanding malignant behavior of OSCC.
