Primary osteoporosis (POP): a systemic skeletal disease characterized by low bone mass and microstructure deterioration of bone tissue, with a consequent increase in bone fragility and susceptibility to fracture risk, is the most common generalized disorder of bone and affects hundreds and millions of women and men in worldwide. But it involves more women than men and its morbidity varies with age. The routine treatment of POP is hormone replace therapy (HRT), for the causes of POP may partly attribute to estrogen deficiency. But many researches had illustrated that estrogen could induce many side-effects if it had been used for long time. So it is a urgent task for medical scientists to find out some drugs that is safe and effective to POP patient. ObjectiveThe study was designed to monitor the dose-effect response of Chinese kidney-tonifying drugs (CKD) on POP animal model, which was established by removing ovary. Then we can assess the effect and explore the mechanism of CKD on the ovariectomized SD rats. Therefore to provide a safe and economical drug that was effective in treating POP. Methods1. Collecting and screening CKD is based on the experiential anagraph on POP and the counsel of specialists. 18 drugs were enrolled, then to formulate Chinese kidney-tonifying drugs .2. 60 female SD rats (six months ) were randomly divided into 6 groups of 10each: sham group, OVX group, OVX+CKD-H group; OVX+CKD-M group; OVX+CKD-L group; OVX+E2 group. All the rats had been ovariectomied except those in sham group, and treated for 3 months separately except OVX group.3. After treatment, we compared the value of bone mineral density (BMD) measured by dual energy X-ray absorptionrnetry (DEXA) between OVX group and sham group, to determine whether the model of POP had been establish successfully or not.4. Effects of estrogen and different dosage levels of CKD on the POP rats were assessed by measuring BMD, histomorphometry indexes, biomechanics indexes, and making ANOVA analysis.5. To explore the mechanism of CKD in curing POP , we measured serum ? and ALP level, urine HOP level, and made ANOVA analysis.6. To observe the side-effect of CKD. Results1. After ovary excised for 3 months, the BMD in rats of OVX group were significantly lower than that of sham group ( p<0.05 ). It indicated that the animal model of POP after ovariectomized had been establish successfully.2. Compared with OVX group, Chinese kidney-tonifying drugs had the better effect of preventing and treating POP.2.1 The effect on increasing the BMD of ovariectomized rats: CKD-H and CKD-M had obviously increased the BMD of whole body( p<0.05 ). The effect of enhancing BMD by CKD-H and CKD-M was equal to that of E2( P>0.05 ), CKD-L could increase part of BMD (arms, legs and trunk )of ovariectomized rats.2.2 The amendatory effect of the bone minuteness structure in ovariectomized rats: one-third of distal right femurs of the rats were isolated and the structure change of the trabecular was observed with microscope. It showed that the distribution, arrangement, space and thickness of trabecular in CKD-H group and CKD-M group were better than those of OVX group, and were similar to those of sham group and E2 group. Bone histomorphology was analysised by computerized image analysis system, the result showed that the trabecular bone volume ( TBV% ) and meantrabecular thickness value ( MTT ) in CKD-H group and CKD-M group were higher than those of OVX group ( p<0.05 ), there was no significantly difference between CKD-H, CKD-M and E2 group ( p>0.05 ). The TBV% in CKD-L group was higher than that of OVX group (p<0.05 ).2.3 The effect on improving the bone biomechanics of ovariectomized rats: . the index of structural behavior of the left femur, such as ultimate load ( UL ) and ultimate deformation ( UD ), and the index of material behavior of the femur, modulus of elasticity, in CKD-H group and CKD-M group were significantly higher than those of OVX group ( p<0.05 ), there were no significantly difference...
