Objective: To study the relationship between intercellular adhesion molecular-1 (ICAM-1), E-cadherin(E-CD), urokinase-type plasmingen activator (uPA), its receptor uPAR, vascular endothelial growth factor(VEGF), its receptor Flk-1, Flt-1 and tumor cell invasion, metastasis in hepatocellular carcinoma (HCC). Methods: The expressions of ICAM-1, E-CD, VEGF, Flk-1, Flt-1 by irnmunohistochemistry and uPA, uPAR by in situ hybridization were detected in 52 cases of HCC, 42 cases of liver cirrhosis and 23 cases of normal liver tissue. Mircrovessel density (MVD) was detecting by irnmunohistochemistry with CD34 monoclonal antibody in all samples.Result: Compare with their expression in liver cirrhosis and normal liver tissue, the expression of ICAM-1, E-CD, uPA, uPAR and VEGF, Flk-1, Flt-1 in HCC were abnormal: E-CD expression was reduced, others were overexpression significantly. The number of MVD in HCC was increased significantly. In HCC, E-CD expression with metastasis was reduced significantly as well others were increased; the overexpression of uPA, uPAR, VEGF, Flk-1, Flt-1 were associated with capsule invasion; overexpression of ICAM-1 and reduced expression of E-CD were associated with low-grade. Increased number of MVD was associated with HCC metastasis and capsule invasion. The expression of ICAM-1, uPA, uPAR, VEGF, Flk-1 and Flt-1 showed positive correlation with tumor angiogenesis. To correlation in HCC, E-CD showed negative correlation with uPA, uPAR; Flt-1 showed positive correlation with ICAM-1, VEGF and Flk-1, others showed significantly positive correlation each other.ICAM-1, E-CD, uPAR, VEGF and Flk-1 were contributed to discriminate the categories of samples: HCC, liver cirrhosis or normal liver tissue. uPAR, VEGF were contributed to discriminate HCC metastasis or not. Conclusion:1 The abnormal expression of ICAM-l, E-CD was associated with low grade.2 All markers' expression were abnormal in HCC, and they may contribute significantly to HCC invasion and metastasis. Detecting uPAR, VEGF in HCC can discriminate metastasis or not.3 The expression of ICAM-l, uPA, uPAR, VEGF, Flk-1 and Flt-1 showed positive correlation with tumor angiogenesis.
