Object Select chitosan as film carrier, then add aloe vera, epidermal growth factor (EGF)et al to prepare oral composite film. To explore the most optimal combination of various compound and the physical-chemical nature of drug film. The effect of composite chitosan film on rat oral ulcer induced by chemical agent were observed.Methods (1)To prepare the composite film with chitosan ,EGF and aloe vera et al. (2)To determine the physical-chemical nature of this film. (3)To assess the effect of composite film on experimental oral ulcer.Result (1) The thickness of composite film was about 0.5mm and drug was well-distributed. The film looked like red-brown and its adherence was good. If it contacted with water after 2 minutes, it would become glue-confection and adhere mucosa strongly. It contained 0.007 gram chitosan, 0.051 gram aloe vera, 40IU EGF, 0.553mg dincaine per cm2 film. The PH of 1% water solution of composite filmwas 6.8±0.1 at 25℃. The melting time in water was about 60-90 minutes. (2)Composite film had good releasing capacity-could release most drug in 2-2.5 hours. (3)The oral ulcer size treated with composite film was smaller than controlled group(p<0.05); average healing time was shorter than other groups(p<0.05); the degree of edema and blodshot were less serious than controlled group; the amount of inflammatory cells was less than controlled group in variousstage (p<0.05); the amount of fibroblast was more than controlled group (p<0.05); the positive cells of PCNA were obvious different from controlled group(p<0.05).Conclusion The result indicated that water-soluble chitin was a kind ofgood mucal carrier material, and adding aloe, EGF et al did not affect its adherence; further its drug release was good and it could guarantee efficient release. The result from rat oral ulcer treated with oral composite chitisan film also suggested it could accelerate the healing of experimental oral ulcer and it had a good development prospect. However, the effective mechanism of this composite film on oral ulcer was still unclear and needed to study in the future. |
