The dysfunction of apoptosis is involved in the pathogenesis of autoimmune diseases. To detect the expression of apoptosis gene Bcl-2 family protein Bcl-2,Mcl-1,Bd-XL and Bax in autoimmune thyroid disease (ATTD) Hashimoto's thyroiditis(HT) and Graves Disease(GD) to explore how they involved in the pathogenesis and developmentMethods Get 61 specimen from thyroid operation, including 33 with HT and 28 with GD.Get 20 specimen form normal thyroid constitution beside thyroid cancer as normal control group. Expression and distribution of Bcl-2, Mcl-1, Bax and Bcl-XL proteins in the specimen from 33 patients with HT,28 patients with GD and 20 controls were investigated with immunohistochemical Elivasin?methods. When cell plasma was buffy in Bcl-2, Bax and Bcl-XL, it's positive, and when nucleus of Mcl-1 was buffy, it's positive. Apply double blind procedure to evaluate the intensity and quantity of experimental sheets.Results The expression of Bcl-2 protein is stronger than other proteins. There is significant statistics difference among the 3 groups (p<0.005). The expression intensity of Bcl-2 among 3 group: GD group> control group> HT group. In HT, it's lower near the area of lymphocytes infiltrating and higher far away the area, it's lower in germinal center of lymphatic follicle and higher in the edge of lymphatic follicle. The expression of Mcl-1 protein is relative lower than Bcl-2 protein. There is a significant statistics difference among the 3 groups (p<0.005). The expression intensity of Mcl-1 among 3 group: GD group> control group> HT group. In HT, it's higher near the area of lymphocytes infiltrating and lower far away the area, it' s higher in germinal center of lymphatic follicle and lower in the edge of lymphatic follicle. The expression of Baxprotein has significant statistics difference among the 3 groups (p<0.005). The expression intensity of Bax among 3 group: HT group> control group and GD group, and there is no statistics difference between GD group and control group. In HT, it's higher near the area of lymphocytes infiltrating and lower far away the area, it' s higher in germinal center of lymphatic follicle and lower in the edge of lymphatic follicle. Expression of Bcl- XL protein in thyroid follicles cells from GD group and HT group is stronger than control group, and there is no statistical difference between GD group and HT group.Conclusion The reduced expression of antiapoptosis proteins Bcl-2 and Mcl-1 in HT is related with high apoptosis rate of thyrote in HT; The increased expression of antiapoptosis proteins Bcl-2 and Mcl-1 has a significance for the increased rates of cell proliferation in Graves disease. The increased expression of pro-apoptosis protein Bax is related with high apoptosis rate of thyrote in HT. The ratio of Bcl-2/Bax is important to cell apoptosis.
