Objective: To study the significance of CD44 and CD54 expression in animal models and patients with liver metastasis of colorectal cancer ,and to find a stable biological marker for the early diagnosis of liver metastasis of colorectal cancer.Methods : Experimental study: fouty- two nude mice of 4-6wks were adopted in the study. Thirty- six were used to establish animal models with liver metastasis of colorectal cancer, the rest 6 mice were used as control. 36 mice of the experimental group were divided into 6 subgroups, each containing 6 mice, which were injected in the subcapsule of the spleen with low grade colorectal adenocarcinoma cells (LS174T).On day 10,15,20,30 and the brink of death, each subgroup were sacrificed by eyeball removal and blood letting. Serum CD44, CD54 and CEA levels of the nude mice were measured by ELISA and the sizes and number of liver metastatic loci were observed and noted. The 6 mice for control were raised in the same cages and were killed on day 30, and were compared with those of the experimental group. In clinical study:Thirty-eight patients with colorectal cancer, 21 patients with liver metastasis of colorectal cancer and 40 healthy controls were enrolled in the study. 3ml of fasting blood was drawn before and after operation (group of colorectalcancer) and after chemotherapy and radiotherapy (group of liver metastas of colorectal cancer ),which then underwent ELISA analysis for CD44,CD54 and CEA.Thelevels of CD44, CD54 and CEA in patients colorectal cancer and liver metastasis of colorectal cancer before and after treatment were compared.,were compared with the normal controls.Results: Through the measurement ,at different phrases, of serum CD44 ,CD54 and CEA level of animal models with liver metastasis of colorectal cancer ,we found with the development progression and metastasis of cancer,CD44 and CD54 levels in the animal serum tend increase gradually,as correlate positively with the growth time ,size,and extent metastasis of the cancer.Through the measurement of serum CD44 and CD54 before and after treatment of patients with colorectal cancer with or without liver metastasis,we further confirmed that CD44 andCD54 have a close positive correlation with the development progression of colorectal cancer.CD44 and CD54 are early tumor markers for liver metastasis of colorectal cancer.Combined detection of 2 markers shows high sensitivity than the detection of CEA alone and may lead to increased early diagnost rates of liver metastasis of colorectal cancer.Conclusions: CD44 and CD54 can act as biological markers for early clinical diagnose of liver metastasis of colorectal cancer.Combmed detection of the two markers has high sensitivity then the detection of CEA alone. CD44 and CD54 also act as markers for forecast after treatment of colorectal cancer and liver metastasis of colorectal cancer.
