Objective: To study the effect and mechanism of VitC, VitE on the blood system and hippocampus of lead poisoning SD rats. Methods:Preventive effect of Vitmin C, Vitamin E on blood system of lead exposed SD rats in development phase25 male SD rats were divided randomly into 5 groups. Blank group, eat and drink freely, lead control group, drink 0. 02% lead acetate (mixed by deionized water) for four weeks. Preventive groups, fed by vitamin C 100mg/kg. bw, vitamin E 100mg/kg. bw, or vitamin C (100mg/kg. bw ) and vitamin E (100mg/kg. bw)respectively at the same time of drinking 0.02% lead acetate. At week 4, concentration of Fe, Zn, Ca, Pb, Hb and -ALAD in blood were measured.Therapeutical effect of Vitmin C, Vitamin E on lead exposed SD rats in development phase25 male SD rats were divided randomly into 5 groups, therapy groups, fed by vitamin C 100mg/kg. bw, vitamin E lOOmg/kg. bw, or vitamin C and vitamin E respectively for one week, after drinking 0. 02% lead acetate for 4 weeks. At week 5, all rats are sacrificed by luxation of cervical vertebra, concentration of Fe, Zn, Ca, Pb, Hb and 8 -ALAD in blood, and activity of SOD, GPx, MDA, NOS and NO of hippocampus were measured. Results: Preventive effect of Vitmin C, Vitamin E on blood systemBlood -ALAD, Kb, Zinc concentration of preventive groups increased significantly than that of lead control group (p<0. 05), but concentration of Pb was lower than lead control gfoup(P<0.05) .serum calciunu zinc level was no significance between these groups (p>0.05).Therapeutical effect of Vitmin C^ Vitamin E on blood system At week 5, concentration of Pb in therapy groups were lower than that of lead control group significantly (p<0. 05). Concentration of Fe, Hb, 8 -ALAD were higher than lead control group significantly(p<0.05). Between all therapy groups and lead control group, no significance was observed about concentration of Ca, Zn (p>0. 05).Protection of Vitamin C, Vitamin E on hippocampus As MDA, its concentration of therapy groups were higher than lead control group significantly(p<0. 05), and among therapy groups, concentration of MDA in combination of VitC and VitE group was higher than VitC therapy groups significantly(p<0.05). As SOD, GPx, NO, NOS, their concentration were higher than lead control group significantly(p<0. 05). Conclusion:Supplied Prophylactically or treat with Vitamin E^ Vitamin C can low blood lead level, increase Fe level, recover activity of ALAD and Hb, decrease injury of blood system by lead. In addition, VitC and VitE can protect hippocampus from Lipid peroxidation, and inversio NO, NOS in lead poisoing rats.
