Objective The purpose is to study the differences in the survival and the percentage of apoptosis between bronchoalveolar lavage fluid (BALF) and peripheral blood (PB),the effect of interleukin-5(IL-5) on eosinophils (EOS) survival. Methods The animal model of asthma was established by OVA sensitized. EOS was insolated form BALF and PB by gradient centrifugation with percoll, then cultured with RPMI 1640 medium. IL-5 (10u/ml) was added into both BALF-EOS and PB-EOS, the reference group was without IL-5. The survival rate and the percentage of apoptosis of two type of EOS were observed at Oh , 24h, 48h and 72h respectively with Arcidine orange (A0)/ Ethidium Bromide (EB) fluorescent staining assay. Results (1)in asthmatic guinea pig model there was no difference of survival rate and the percentage of apoptosis at Oh cultured between BALF-EOS and PB-EOS (P>0. 05). (2) The percentage of apoptosis of cultured PB-EOS were significantly higher than that of cultured BALF-EOS at 24h, 48h and 72h (P <0. 05). The survival rate of the cultured PB-EOS was significantly lower than that of cultured BALF-EOS at 24h, 48h and 72h (P<0. 05). (3) The percentage of apoptosis cells of the PB-EOS group with IL-5 was significantly lower than that of PB-EOS group without IL-5 at 24h, 48h and 72h (P<0. 05) . The survival rate of the PB-EOS group with IL-5 was significantly higher than that of PB-EOS group without IL-5 at 24h, 48h and 72h (P <0.05). (4)The percentage of apoptosis of the BALF-EOS group with IL-5 was significantly lower than that of BALF-EOS group without IL-5 at 24h, 48h and 72h (P <0. 05). The survival rate of the BALF-EOS group with IL-5 was significantly higher than that of BALF-EOS group without IL-5 at 24h,48h and 72h (P <0.05). (5) The percentage of apoptosis of the BALF-EOS group with IL-5 was significantly lower than that of PB-EOSgroup with IL-5 at 24h, 48h and 72h (P <0. 05) . The survival rate of the BALF-EOS group with IL-5 were significantly higher than that of PB-EOS with IL-5 at 24h, 48h and 72h (P <0. 05). Conclusion 1. In asthmatic guinea pig model there was no differences in the percentage of apoptosis and the survival rate at Oh cultured between BALF-EOS and PB-EOS. 2. The percentage of apoptosis of the BALF-EOS was lower than that of PB-EOS at 24h, 48h and 72h (P<0. 05). The survival rate of the PB-EOS was significantly lower than that of BALF-EOS at 24h , 48h and 72h. 3. IL-5 was able to prolong the survival of both PB-EOS and BALF-EOS. But it had a stronger effect on BAL-EOS.
