| Objective To study changes of the messenger RNA expression of collagen types I , XII and transforming growth factor -β1 in keloids after treatment withTetramethylpyrazine(TMP) , and to explore its therapeutic effect and molecular mechanism.Methods There were twenty-two samples including 12 cases of keloid , 10 cases of normal skin tissue. Self-control and normal skin control were used,. Six keloids were treated by injiection of TMP locally for 3 months. And then total RNA was isolated from the tissue, furthermore the messenger RNA expression of collagen I , XII and transforming growth factor -β1 were detected and quantitated with reverse transcription-polymerase chain reaction (RT-PCR).Results Among these cases, after TMP treatment ,downregulation of type I collagen mRNA expression was observed, and there was no statistical difference between the treated group and the normal skin group. The expression of collagen XH mRNA level increased compared with the untreated group, and both the treated group and the untreated groupexpress more than normal skin group. The expression of TGF -β1 mRNA level in the treated group decreased significantly, which is as much as the normal skin tissue.Conclusion The elevated amount of type I collagen antagonizes by TMP with changes of the collagenous composition in tissues, in part by the regulation of cytokines. It is thought that the TMP is valuable for treating keloids. |
