Objective: To observe the expression time course of tumor necrosis factor and interlukine-8 in serum and spinal cord tissue after acute spinal cord injury. To study whether methylprednisolone intervention after acute spinal cord injury has an ability to inhibit tumor necrosis factor alpha and interlukine-8 or not.Methods: Spinal cord injury models were created in 72 adult Wistar rats with improved Allen's weight drop methods(50g.cm).The animals were randomly divided into three groups:control group, SCI group, MP group. In each group ,TNF- and IL-8 were detected in serum and spinal cord tissue with radioimmunoassay respectively .And immunohistochemistical staining were performed in SCI group and MP group. In each group,the optical density of TNF- a positive cells in the adjacent sections of the injured site were measured by image pattern analysis.Results: In the site of injured spinal cord ,the expression of TNF- a and IL-8 up-regulated remarkably .TNF- a up-regulated after SCI 1h ,peaked at 12h .Although it decreased after SCI 24h, the level of TNF- in the cord was 4.3 fold against control group at SCI 72h. IL-8 up-regulated after SCI 3h ,kept increasing for 72h.There was a prominent increase in immunotainning of TNF- in traumatized spinal cord.TNF- and IL-8 could also be detected in serum, and there was a significant increase Methylprednisolone inhibited TNF- a and IL-8 significantly in earlier phases.But after SCI 72h,the level of TNF- a didn't decrease significantly any more. Conclusion: TNF- a and IL-8 increase after SCI,the over expressed TNF- a and IL-8 may contribute to acute spinal cord injury. Methylprednisolone intervention after acute spinal cord injury may attribute to inhibiting these two cytokines partly and further toinhibiting inflammation. Methylprednisolone should be used as early as possible.And the effect of methylprednisolone is limited, methylprednisolone intervention combined with other comprehensive treatment for SCI is necessary.
