| Background & Aims: In recent years, some studies have suggested that salvia miltiorrhiza Bunge (SmB) have perfect anti-fibrotic and some extent anti-portal hypertensive (PHT) effect. Calcium channel blocker (CCB) also has some anti-fibrosis effect. It has been used for portal hypertension and esophageal varices. In this study, we investigated the effects of the early and long-term administration of SmB or verapanil (Ver) alone or combination in rats with carbon tetrachloride (CCL4)/ ethanol induced cirrhosis.Methods: 80 wistar rats were divided into five groups: nomal control group, liver cirrhosis model group. Salvia miltiorrhiza group, Verapanil group, double-drug group. 16 rats in every group The verapanil group was given drug suspension by oral administration every other day. The salvia miltiorrhiza group was given drug by abdominocentesis every day. While double-drug group were given two drugs the same time. In the meantime, the rats were injected CCL4 hypodermically every four days. At the fourth and eighth week, six rats from each group were selected. Portal vein pressure (PVP) were measured and the parameters of serum ALT. hyaluronic acid (HA). Type IV -Collagen (IV-C) and histogical staining of hepatic tissue were evaluated.Results: SmB and Ver can decrease the level of PVP, ALT, HA, IV-C, Double drug have even better effect. In histopathology, two drugs can reducethe hepatocell degeneration and nectosis and delay the formation of pseudo lobe.Conclusions: SmB plus Ver significantly decreased the extent of hepatic fibrosis and significantly reduced the portal hypertension in rats with CCL4/ethanol induced cirrhosis. |
