Objective: To evaluate the effects of polymorphisms of TNF-α-238, -308 in peripheral blood mononuclear cells (PBMCs) on susceptibility to HBV and prognosis of patients infected with HBV.Methods: Using PCR, dots blot and DNA sequencing to study the polymorphisms of TNF-α-238, -308 in PBMCs of patients with HBV infection(n=106) and controls(n=23).Results: There was no significant difference between dots blot and DNA sequencing (p>0.05). No -238A/A positive case was detected in all the samples using dots blot, 3.9% of -238A/G cases were found, 3.9% of -308A/G and 33.3% of-308 G/G in all the samples. There was no significant difference of distribution of-238A/G in each group (p>0.05). But between liver cirrhosis, chronic hepatitis and carriers it had statistical significant (p<0.05). To the distribution of-308 A/A and A/G, there was no significant difference between each group (p>0.05).Conclusions: The results of dots blot and DNA sequencing were coherent. The polymorphisms of TNF-α-238, -308 were not related with HBV infection and polymorphisms of TNF-α -308 was not associated with the extent of diseases, however the incidence of liver cirrhosis increased in patients with TNF-α-238G/G if the person was infected with HBV.
