The Expression Of PTEN, Bcl-2, Bax Oncoprotein As Well As Proliferationactivity And Apoptosis In Non-small Cell Lung Cancer And Their Correlation With Clinicopathological Features And Prognosis | | Posted on:2005-12-02 | Degree:Master | Type:Thesis | | Country:China | Candidate:W C Yu | Full Text:PDF | | GTID:2144360122998012 | Subject:Internal Medicine | | Abstract/Summary: | PDF Full Text Request | | Background The uncontrol Of cell cycle is an impotant reason of carcinomas.Cell is in balance of proliferationactivity and apoptosis and cell proliferationactivity and apoptosis are regulated by gene.When the gene of proliferationactivity overexpress or the gene of suppressed proliferationactivity mutate or lack and the gene of apoptosis mutate and lack or the gene of suppressed apoptosis overexpress,the balance of proliferationactivity and apoptosis is broken,which induces carcinomas.The studies of oncogene and suppressor show that the expression of gene of carcinomas is different and complex.lt is very difficult to confirm which or more genes play very important role in carcinomas.But it is confirmed that the regulation of gene of proliferationactivity and apoptosis plays an important role in developing carcinomas.The gene therapy in carcinomas has developed very fast,and the gene therapy has been used in the clinic from gene studies.Several plans of gene theyapy in the clinic were approved by FDA.Which gene is the best gene in the gene therapy of lung cancer,that is an important question in gene therapy as well as the aim of our study.Objective To study the expression of PTEN,bcI-2 and bax as well as their relationship with proliferation activity and apoptosis in non-small cell lung cancer(NSCLC), and to evaluate the clinicopathological features and clinical usefulness of the correlation.Methods Apply the immunohistochemistry stain (SABC) method to investigate the expression of PTEN> bcl-2 .. bax protein and PCNA in archival wax-embedded specimens of 43 NSCLC and 20 normal lung tissues adjacent to lung carcinoma, TUNEL technique was applied to detect the apoptosis .The apoptosis index(AI) and mitotic index(MI) were defined.Results The positive rate of PTEN is significantly lower in patient with NSCLC than that in normal lung tissues adjacent to carcinomas(46.51%% versus 95%, P< 0.05). The expression of PTEN had no relationship with clinicopathological factors including age, sex, tumor size, TNM stage, histological type of lung cancer .The positive rate of PTEN was higher in high differentiation group than in low differentiation group ,and in lymphnon-metastasis group than in lymph node metastasis group ,and in survival group than in dead group. The positive expression rate of bcl-2 is significantly higher in NSCLC than that in normal lung tissues adjacent to carcinomas(51.16 % versus 20%, P<0.05);No relationship was found among the expression of bcl-2 and clinicopathological factors such as sex, age, tumor size,differentiation degree, histoclassification of lung carcinoma ,lymph node metastasis and prognosis of the cancer cases.The expression of bcl-2 was lower in patient in stage III-IV group than that in stage I~ II group .The positive rate of bax is significantly lower in patient with NSCLC than that in normal lung tissues adjacent to carcinomas(60.47% versus 90%, P< 0.05). The expression of bax had no relationship with clinicopathological factors including age, sex, tumor size, TNM stage, lymph node metastasis and histological type of lung cancer .The positive rate of bax was higher in high differentiation group than in low differentiation group , and in survival group than in dead group. The AI of cancer cases with PTEN-positive was obviously higher than that with PTEN-negative, and so is the AI of the cancer cases with bax -positive , The AI of cancer cases with bcl-2-positive was obviously lower than that with bcl-2-negative. The AI of cancer cases was higher than that of normal lung tissues adjacent to lung carcinoma. No relationship was found among AI and clinicopathological factors such as TNM stage, lymph node metastasis and histoclassification of lung carcinoma But AI had correlation with differentiation degree and prognosis of the cancer cases. The MI of cancer cases with PTEN-positive was obviously lower than that with PTEN-negative , but there was no relationship between MI and the expression of bcl-2 , bax oncoprotein.
