Objective: To study the expression and clinic significance of PTEN,BCL-2 and C-erbB-2 protein in endometrial carcinoma and its relationships. Methods: Using immunohistochemistry test (s-p methods) , We examined the expression of PTEN,BCL-2 and C-erbB-2 protein in 50 cases of endometrial carcinoma , 20 cases of endometrial precancerous lesions and 20 cases of normal endometrium. Results: (1)In endometrial carcinoma tissues , the positive rates of PTEN protein expression and BCL-2 protein expression were significantly lower than that of uterus atypical hyperplasial endometrium and normal endometrium tissues (X~2 = 15.271 , X~2 = 14.081 , P<0.05) ,but the positive rates of C-erbB-2 protein expression were significantly higher than that of uterus atypical hyperplasial endometrium and normal endometrium tissues (X~2 = 12.482 , P<0.05). (2)The expression of PTEN protein correlated with histological grade(X~2 = 6.317 , P<0.05) ,muscles invasion(X~2 = 5.556 , P<0.05),clinical stage (X~2 = 6.523 , P<0.05) and pathological type(X~2 = 5.357 , P<0.05) , but not with lymph node metastasis (Fisher'Exact test, P > 0. 05) .In endometrial carcinoma , with progression of clinical stage , increased histologic grade and deepening of muscles invasion , the positive rate of PTEN protein expression was correspondingly decreased. In endometrioid adenocarcinoma , the expression of PTEN protein was higher than that in other pathological type. (3) The expression of BCL-2 protein in human endometrial carcinoma was related to histological grade (X2 = 6.538 , P<0.05),muscles invasion(X2 = 10.470, P <0. 05) and clinical stage (X2 = 6.190 , P<0.05) , but not with lymph node metastasis (Fisher'Exact test,P>0.05) and pathological type (X2 = 1.176 , P >0. 05).In endometrial carcinoma , with progression of clinical stage , increased histologic grade and deepening of muscles invasion , the positive rate of BCL-2 protein expression was correspondingly decreased. (4)The overexpression of C-erbB-2 protein was correlated with muscles invasion(X2 = 5.362 , P<0.05) and clinical stage (X2= 8.696 , P<0.05). In endometrial carcinoma , with progression of clinical stage , and deepening of muscles invasion , the positive rate of C-erbB-2 protein expression was correspondingly decreased. With increased histologic grade , the potential of C-erbB-2 protein expression was risen, but there was no difference in statistics(X2 = 4.084 , P>0.05)。Neither lymph node metastasis (Fisher'Exact test,P>0.05) nor pathological type (Fisher'Exact test,P>0.05) correlated with the expression of C-erbB-2 protein. (5) The expression of PTEN protein were positively correlated with BCL-2 protein (Rs= 0. 850 , P < 0.001) , but were negatively correlated with C-erbB-2 protein in endometrial carcinoma (Rs=-0.879, P < 0.001) . The expression of BCL-2 protein were negatively correlated with the expression of C-erbB-2 protein. Conclusion : The results suggest that absent PTEN,BCL-2 and C-erbB-2 protein overexpression may play an important role in the genesis and development of endometrial carcinoma. The expression of PTEN protein was strongly associated with the expression of abnormal BCL-2 and C-erbB-2 protein. There may be a synergetic action among these genes .Binded detections of PTEN,BCL-2 and C-erbB-2 protein have positive effect onearly diagnosis , prognosis assessment of endometrial carcinoma, and provide clinic treatment with indicators.
