Objective To explore the effect of grape procyanidins(GPC) on hyperplasia activity of the vein blood lymphocytes of humanbody.(1) To study the effect of GPC on cell proliferation of the normal lymphocytes of humanbody.(2) To study the protective effect of radiation injury of GPC on hyperplasia activity of the lymphocytes of humanbody. .(3) To study the effect of GPC on cell proliferation of the lymphocytes of radiatherapy patient.Method The vein blood samples were taken from 21 healthy young women and 5 radiotherapy patients. Each sample was divided into three groups, which were high-dose GPC-treated group, low-dose GPC-treated group and control. (1) The vein blood of healthy young women were given GPC, and after cultured 24 and 48 hours, the lymphocyte apoptosis rate was measured by flow cytometry; after cultured 48 hours, the lymphocyte proliferation was measured by MTT assay; after cultured 72 hours, the lymphocyte transfermation rate was measured by morphotogical method. (2) The vein blood of healthy young women were vitro irradiated by X-ray; with the electron liner acceleyator. and then given GPC, and also ,after cultured 24 and 48 hours, the lymphocyte apoptosis rate was measured by flow cytometry; after cultured 48 hours, the lymphocyte proliferation was measured by MTT assay; after cultured 72 hours, the lymphocyte transfermation rate was measured by morphotogical method. (3)After cultured 24 and 48 hours, the lymphocyte proliferation of the vein blood of the radiotherapy patients was measured by MTT assay.Result (1)the normal samples: The levels of lymphocty apoptosis rate, cell proliferation and transfermation rate of high-dose GPC-treated group were17.18?.19, 0.31+0.02, 0.88 + 0.03; and of normal controls were 29.51 ?4.91, 0.19?.2, 0.72?.03, Compared to the normal controls, the levels of apoptosis rate of high-dose GPC-treated groups were lower, the levels of cell proliferation and transfermation rate were both higher. All the differences above were stastiscally significant (P<0.01). (2)the normal irradiated samples: The levels of lymphocty apoptosis rate, cell proliferation and transfermation rate of high-dose GPC-treated group were 26.24?. 04, 0.22?. 02, 0.84?0.06; and of positive controls were 38.24?. 99, 0.16 + 0.02, 0.64?.04., In high-dose GPC-treated groups, the levels of apoptosis rate were lower, and the levels of cell proliferation and transfermation rate were higher than the levels of positive controls. All the differences above were stastiscally significant (P<0.01). (3)the radiothererapy patient samples: After cultured 24 hours, the levels of high-dose GPC-treated group, low-dose GPC-treated group and control were 0.27+ 0.01,0.18?0.02,and 0.1 8 ?0.01. The levels of high-dose GPC-treated group were much higher than the levels of controls. The difference was stastiscally significant (p<0.01).Conclusion (1) GPC can increase the cell proliferation of the normal lymphocytes.(2) GPC has a significant protective effect on cell proliferation of radiation injured lymphocyts.(3)GPC can improve the cell proliferation of the lymphocytes of. the radiothererapy patient.
