In the support of the National Natural Science Foundation, we applied Didangtang as the representative of Blood-activating and stasis-eliminating herbs to interfere with the model on CT-26 cell spleen transplant and liver metastasis of mice colorectal cancer. So that we can observe the effect of Didangtang on tumor metastasis, NF-kB and EGFR in the mice model and discuss the effect and mechanism of Blood-activating and stasis-eliminating herbs on tumor metastasis.Objective: To study the effect and mechanism of Didangtang on tumor metastasis, which is the representative of Blood-activating and stasis-eliminating herbs.Mehtods: Forty-eight BALB/C mice were randomly divided into four groups: The normal group, the model group, Didangtang group and Danshen group. Except for the normal group, the other three groups were carried out the operation of CT-26 cell spleen transplant and liver metastasis of colorectal cancer and no treatment was carried out on the normal control group. After the operation, the Didangtang group is treated with Didangtang gastric clysis; the Danshen group was treated with Danshen gastric clysis; while the model group and the normal control group were both treated with physiological brine gastric clysis. Two weeks later killed the mice and studied the cytological and morphologic changes of hepatocytes which were treated with immunohistochemical SP method, Two-step method coloration and pathological HE coloration.Results: 1.Compared with the normal control group, there was remarkable difference (liver weight P<0.01, tumor nodes amount P<0.01) in the liver weight and tumor nodes amount among the three groups inoculated tumor cells. The liver weight of Didangtang group was lighter than that of the model group (P<0.05) and Danshen group(P<0.01). The tumor nodes amount of Didangtang group was less than those of the model group (P<0.05) andDanshen group (P<0.01) too. While there was no remarkable significance in terms of liver weight and tumor nodes amount between model group and Danshen group (liver weight 0.750.1). So we considered the positive rate of the Didangtang group was lower than that of model group . As far as positive rate of the liver EGFR of Didangtang group was concerned, when compared with that of the normal control group, there was remarkable difference (x2=5.282,P<0.01); when compared with that of Danshen group, there was no remarkable difference(x2=1.173,P>0.1). It indicates that Didangtang can obviously inhibit the expression of EGFR. 4.There was remarkable difference(x2=6.362,P<0.05) among the liver NF-kB positive rate of the three operated groups.
