Objective To investigate the expression of transforming growth factor -beta-l(TGF-β1) and basic fibroblast growth factor (bFGF) and their relationships with cellular proliferation in human glioma.Methods The expression levels of TGF-β1,bFGF and proliferative cell nuclear antigen (PCNA) in 35 cases of human glioma and 7 cases of normal brain tissue specimen were detected by immuno- histochemical technique. Furthermore, the correlation among the expression of the TGF-Pi ,bFGF and PCNA was compared.Results In normal brain tissue, glioma grade I -Hand grade III-IV, the positive expression level of TGF-β1 was 6.57±1.72,13.75 ±4.14 and 36.07±10.13 respectively ,and the level of bFGF was 9.29±2.69, 23.85±7.76 and 42.53±12.37 respectively, and the level of PCNA was 3.86±1.07,24.15±6.12 and 57.07±17.26 respectively .The differences were all significant (p<0.05)in normal brain tissue , glioma grade I - II and grade III-IV. In 35 cases of human glioma and 7 cases of normal brain tissue specimen ,there was a postive correlation between TGF-β1 and bFGF level (r=0.79, P<0.01),a postive correlation between TGF-β1 and PCNA level (r=0.73, P<0.01), and also a postive correlation between bFGF and PCNA level (r=0.74, P<0.01).Conclusion There is an intrinsic correlation between the expression of the TGF-β1,bFGF and cellular proliferation and histological grade of human brain gliomas . These findings suggest that the synergistic interaction of TGF-β1 and bFGF play an important role during the cellular proliferation of glioma.
