| Male reproductive system can also secrete estrogen which conduct its biological effects through combination with estrogen receptor. In our lab, the formal study hasused immunocytochemistry to localize the estrogen receptor- a in the efferent ducts.Anatomically, the efferent ducts extend from the rete region of the testis to the initial segment of the epididymis. The major role of the efferent duct is fluid resorption. Dysfunction of the efferent ducts leads to the impairment of male fertility. 1. The present study has used immunocytochemistry to localize a water channel protein, Aquaporin-l(AQP-l), to the efferent ducts of male adult Wistar rats. 2. Rat were administered DES neonatally. we observed the whole weight of two testis with epididymis in DES-treated rats. We observed the efferent ducts lumen and the epithelial cell height in the efferent ducts used HE method in DES-treated rats .We observed the immunoexpression of AQP-1 using immunocytoshemistry method in DES-treated rats. The comparison of the weight of testis, the epithelial cell height and the mean intensity of AQP-1 obtained using Scion Image for Window was made using SPSS 11.0 for window .The results show: 1. In the efferent ducts, AQP-1 was expressed on the microvilli,basolateral plasma membranes, and apical endosomes of the nonciliated cells and cilia of ciliated cells.2.the weight of testis was not influenced at age 10 days, but influenced at other days in DES-treated rats. The weight of testis was reduced 55% at age 18 days,47% at age 25 days, 80%at age 35 days and 60 days. We can recognized the efferent ducts with the irregular lumen using HE method. In DES-treated rats, we couldn't recognized well the efferent ducts because their lumen became regular using HE method. The efferent ducts lumen became regular because the efferent ducts were diluted and the epithelial cell height was shortened. But we could recognize the efferent ducts because the cilia of the ciliated cells became red using HE staining .The epithelial cell height became higher and higher with time passing by. But the epithelial cell height of efferent ducts in DES-treated rats growed up slowly . At all stages, the epithelial cell height of efferent ducts in the control ratswas higher than that in the tats in DES-treated rats. The epithelial cell height of efferent ducts in DES-treated rats was reduced 45% at age 10 days, 41% at age 18 days, 37% at age 25 days, 71% at age 35 days and 69% at age 60 days .Using immunocytochemistry method , the immunoexpression of AQP-1 was similar at all stage in control rats . But AQP-1 staining was reduced at all stage in DBS-treated rats. The mean intensity was reduced 40% at age 10 days, 36% at age 18 days, 42% at age 25 days, 32% at age 35 days and 30% at age 60 days. The AQP-1 staining was similar at all stages in DBS-treated rats .Conclusion: 1. These results suggest that AQP-1 plays an important role in the fluid reabsorption in the efferent ducts . 2. Injected estrogen can damage the fluid reabsorption in the efferent ducts, which could lead to the impairment of male fertility . |
